2种测定N-乙酰半胱氨酸含量的方法比较

【摘要】 目的:比较2种测定N-乙酰半胱氨酸(NAC)原料含量的方法。方法:采用容量法和高效液相色谱(HPLC)法对NAC原料进行含量测定。结果:容量法测定结果偏高,HPLC法NAC检测浓度在0.1080~1.2690mg·mL-1范围内与峰面积积分值线性关系良好(r=0.9995);低、中、高平均加样回收率分别为99.77%、100.20%、100.20%,RSD=0.48%,含量测定结果较容量法低。结论:容量法测NAC含量操作简单,方法快速;HPLC法能够比较直观、真实的反映样品中所含有的杂质,与容量法比较,含量偏低。

【Abstract】 OBJECTIVE:To compare two different methods for the content determination of acetylcysteine raw material.METHODS:The content of acetylcysteine was determined by titration and HPLC respectively.RESULTS:The result of the titration was higher that that of HPLC method.The standard curve was linear over the range of 0.108 0~1.269 0 mg·mL-1(r=0.999 5) with an average recovery rates of it at low,medium and high concentrations 99.77%,100.20%,100.20%(RSD=0.48%) by HPLC.CONCLUSIONS:The method of titration is simple and quick;but HPLC can intuitively and really reflect the impurities of the sample,compared to titration,the content of samples is lower.
