纤维素酶法制备壳寡糖工艺的研究
Study of Preparation Technique for Chitooligosaccharides by Cellulose
【摘要】 以虾壳壳聚糖为原料,纤维素酶为催化剂制备壳寡糖,探讨了纤维素酶添加量、酶解温度、溶液pH值及酶解时间对壳寡糖得率的影响。通过单因素和正交实验确定酶解最优条件为:纤维素酶添加量1.2g/dL、酶解温度50℃、溶液pH值4.5、酶解时间10h,此条件下壳寡糖得率达到32.15μg/mL;各因素对壳寡糖得率的影响依次为酶解温度>酶解时间>溶液pH值>纤维素酶添加量。
【Abstract】 Chitooligosaccharides are prepared from chitosan of shrimp shell by cellulose.The effects of four reaction parameters,including amount of cellulose,enzymolysis temperature,pH and enzymolysis time on the yield of chitooligosaccharides are conducted to explore by single factor experiment and orthogonal experiment.The results show that the optimal condition is cellulose addition amounts of 1.2 g/dL in the condition of 50 ℃ and pH value of 4.5 for 10 hours.Under these conditions,the yield of chitooligosaccharides is 32.15 μg/mL.In addition,enzymolysis temperature has the largest impact on the yield of chitooligosaccharides followed by enzymolysis time,pH value and amount of cellulose.
