甜菜抗草甘膦除草剂基因的克隆
Molecular Cloning of Glyphosate-resistant Gene for Sugarbeet
【摘要】 常规甜菜对除草剂表现为高度敏感。为了获得抗草甘膦除草剂转基因甜菜材料,通过RT-PCR技术,对草甘膦靶基因AtEPSPs(GenBank No.NM202267.1)的功能域进行克隆。将扩增后的片段连接到测序载体pMD 18-T,进行酶切和测序鉴定。结果表明,AtEPSPs基因的核苷酸长度为1470 bp,推断编码490个氨基酸,确认获得了抗草甘膦除草剂基因功能片段。
【Abstract】 Beta vulgaris is extremely sensitive to herbicidal chemicals.Target gene AtEPSPs(GenBank No.NM202267.1) was selected to improve varieties of sugarbeet with glyphosate-resistance,and the CDS of AtEPSPs was cloned by RT-PCR.AtEPSPs was constructed into pMD 18-T vector,and identified with enzymes digested and sequenced.AtEPSPs was 1470 bp,deduced to encode 490 amino acids,and identified as prospection.
