红麻光钝感突变体光周期反应特性与RAPD扩增片段差异分析

【摘要】 为了研究红麻光钝感突变体基因的遗传规律及与红麻光钝感形成有关的基因序列特征。以红麻品种福红952经航天诱变获得的光钝感突变体与光敏感红麻细胞质保持系L23B品种杂交,杂交后代即F2群体在海南130d短日照条件下诱导了花蕾的分化发育,统计其分离情况,同时利用经筛选的多态性较好的RAPD引物扩增光钝感红麻与对照品种基因组DNA。结果表明:光钝感与光敏感性状存在一对基因的遗传差异,红麻光周期钝感性状为隐性遗传。从24个多态性较好的RAPD引物中筛选到3个引物可以把红麻光钝感突变体与对照品种区分开来,其中有1条(1178bp)是编码蛋白的基因片段,说明RAPD不仅扩增基因组上的非编码蛋白序列,同时可扩增编码蛋白的基因片段,反映不同红麻光钝感材料遗传上的差异。该研究结果可为进一步开展红麻光钝感的基因克隆及分子机理研究提供重要的遗传材料。

【Abstract】 This paper aimed at studying the genetic features of kenaf light-insensitive mutant.A light-insensitive mutant which was from Fuhong 952 mutated in aerospace,and a light sensitive cytoplasmic maintainer L23B were crossed as parents,the F2 population was induced to differentiate buds for 130 d under short-day condition in Hainan province,and the genetic separation of the F2 was analyzed.At the same time,the screened polymorphism RAPD primers were used to amplify genomic DNAs of the light insensitive mutant and the CK.The results showed that there existed three phenotypes in F2 generation,and the separation ratio was 3:1 of the light sensitive to the light insensitive ones. Three polymorphic RAPD primers screened from the 24 primers could be distinguished from the CK.A sequence fragment encoding protein was founded,which illustrated that RAPD not only amplified non-protein coding sequences,but also amplified protein coding sequences.This result revealed that there was difference in a pair of genes between light insensitive and light sensitivity kenaf,and the genetic trait of the light insensitive kenaf was recessive.The results might provide important genetic materials for studying light insensitive kenaf gene cloning and molecular mechanism further.
