高强度聚焦超声制备肿瘤疫苗对小鼠抗肿瘤免疫增强作用的研究
Study on Enhancement of Murine Anti-tumor Immunity Induced by High Intensity Focused Ultrasound Prepared Tumor Vaccine
【摘要】 目的探讨高强度聚焦超声(HIFU)制备的肿瘤疫苗对小鼠抗肿瘤免疫的增强作用。方法每组小鼠20只,分别以2种声强的HIFU瘤苗、高温瘤苗及生理盐水注射,接种H22细胞后观察小鼠肿瘤发生率及生存期;检测免疫小鼠淋巴细胞的增殖及小鼠细胞毒性淋巴细胞(CTI)对肿瘤的杀伤;观察不同声强对HIFU瘤苗效能的影响。结果 HIFU瘤苗组小鼠肿瘤发生率减低,生存期延长;淋巴细胞在同源肿瘤刺激下增殖明显,CTL对肿瘤细胞有特异性的杀伤,且比高温瘤苗组有更高的增殖率和杀伤率(P<0.05);显著提高声强后制备的HIFU瘤苗效能降低。结论一定参数下制备的HIFU瘤苗明显增强了小鼠抗同源肿瘤的免疫力。
【Abstract】 Objective To investigate the anti-tumor immunity induced by high intensity focused ultrasound(HT FU) prepared tumor vaccine in mice model.Methods After inoculation with HIFU prepared or heat solidified tumor vaccines or normal saline,20 mice in each group were transplanted with H22 cells.Then occurrence of tumor and life span of all mice were observed.Proliferation of mice lymphocytes stimulated by tumor cell and the kill-rate of cytotoxic T lymphocyte(CTL) against tumor cell were investigated.Different sound intensity of HIFU was used to study its influence on vaccine.Results Mice with HIFU prepared vaccine had lower tumor occurrence and elongated survival. Co-cultured with cognate tumor cells,lymphocyte proliferation and tumor kill-rate were increased,which were more significant than those of mice with heat solidified tumor vaccine.Extremely enhanced sound intensity of HIFU attenuated the vaccine efficiency.Conclusions HIFU prepared tumor vaccine with certain parameters can enhance anti-tumor immunity of model mice.
