雄激素对实验性自身免疫性脑脊髓炎大鼠星形胶质细胞的影响
Effects of exogenous androgen on astrocytes in rat experimental autoimmune encephalomyelitis model
【摘要】 目的研究外源性雄激素对实验性自身免疫性脑脊髓炎大鼠中枢神经系统星形胶质细胞的影响。方法用豚鼠全脊髓匀浆免疫Wistar大鼠诱导建立多发性硬化动物模型实验性自身免疫性脑脊髓炎(experimentalautoimmune encephalomyelitis,EAE),应用胶原纤维酸性蛋白免疫组化的方法观察丙酸睾酮对EAE大鼠星形胶质细胞数量和形态影响的变化。结果 60%的大鼠成功建模;雄激素干预组发病率降低,该组大鼠的大脑、脑干、脊髓星形胶质细胞数量明显较模型组减少,胞体较正常对照组大,但较模型组明显减小(P<0.05)。结论雄激素可减轻EAE中星形胶质细胞反应。
【Abstract】 Objective To observe the effects of exogenous androgen on astrocytes and NKT cells in experimental autoimmune encephalomyelitis(EAE)in Wistar rats.Methods EAE was induced by immunizing with spinal cord homogenate of guinea pig and evaluated by histological staining.Effects of testosterone propionate on the number and the shape of astrocytes were observed by immunostaining of glial fibrillary acidic protein.Results EAE was successfully induced in 60%rats and androgen treatment could reduce the incidence rate.The number and morphology of astrocytes in myelin of cerebrum,brain stem and spinal cord of model group were significantly(P<0.05)different from that in androgen treatment group.Conclusion Exogenous androgen might exert its protective/therapeutic effects in rat EAE model partially through repressing astroglial reaction of central nervous system.
