龙胆苦苷脂质体的制备工艺研究

【摘要】 目的对龙胆苦苷脂质体的处方和制备工艺进行研究。方法以包封率为评价指标,采用逆相蒸发法制备龙胆苦苷脂质体,并对工艺条件进行正交设计以及单因素考察。结果确定龙胆苦苷脂质体工艺条件为大豆卵磷脂与胆固醇用量比2∶1;大豆卵磷脂与龙胆苦苷用量比25∶1;磷酸缓冲液pH 7.0,超声时间10 min,温度40℃;3批样品的平均包封率为69.78%,平均粒径为183 nm。结论逆相蒸发法制备龙胆苦苷脂质体工艺稳定,重复性好。

【Abstract】 AIM To study the formulation and preparation of gentiopicroside liposome.METHODS Evaluated by entrapment efficiency,the gentiopicroside liposomes were prepared by a reverse-phase evaporation technique to establish the conditions of preparation with orthogonal test and single-factor experiment.RESULTS The optimum conditions were as follows: the mass ratio of soya bean lecithin and cholesterol was 2∶ 1.The mass ratio of soya bean lecithin and gentiopicroside was 25∶ 1.Phosphate buffer pH was 7.0.The time of supersonic was 10 min.Temperature was at 40 ℃.The average entrapment rate of three batches of samples was 69.78%,and the average partical size was 183 nm.CONCLUSION The preparation process of gentiopicroside liposome with a reverse-phase evaporation technique is stable and reproducible.
