薄膜封包对慢性皮炎小鼠模型表皮增生的影响

【摘要】 目的探讨薄膜封包对慢性皮炎小鼠模型表皮增生的影响。方法以0.01%佛波酯(TPA)涂搽小鼠皮肤,造慢性皮炎表皮增生模型,应用非通透性膜封包,采用组织病理及免疫组化法观察表皮增生改变及增殖细胞核抗原(PCNA)的表达。结果 TPA保鲜膜组和TPA基质组,表皮厚度(μm)分别为89.94±14.34,84.28±20.64,t=2.51;PCNA阳性标记率(%)分别为20.12±1.04,19.85±1.68,t=0.52,两者比较差异无统计学意义(P>0.05)。结论利用0.01%佛波酯(TPA)反复涂擦小鼠背部可以复制慢性皮炎模型;增殖细胞核抗原在慢性皮炎动物模型中表达明显增高;薄膜封包对慢性皮炎小鼠模型表皮增生没有明显调节作用。

【Abstract】 Objective To study the effect of epidermic proliferation covering with membrane on the mouse model of chronic dermatitis.Methods The model of chronic dermatitis showing the epidermic proliferation was produced by repeated application of 0.01%TPA on the mouse flanks.The mouse flanks was covered with the airtight membrane in order to chang the epidermic barrier function and the chang of epidermic proliferation was observed by the method of tissue pathology and expression of proliferation cell nuclear antigen.Results Epidermal tickness were(89.94±14.34、84.28±20.64μm,t=2.510)and expression of proliferation cell nuclear antigen were(20.12±1.04、19.85±1.68%,t=0.52) between the model of TPA-control and TPA-airtight membrane group(P>0.05),There were no statistical differences between the two group.Conclusion Repeated application of 0.01%TPA on the mouse back can copy the models of epidermal proliferation in chronic dermatitis.The level of PCNA expression was increased obviously in the model of chronic dermatitis.Covering with memberance on the mouse model of chronic dermatitis can not effectively adjust the epidermic proliferation in the model of chronic dermatitis.
