外源性甲状腺素对梗阻性黄疸大鼠肝脏纤维化的保护作用研究

【摘要】 目的探讨外源性甲状腺素对梗阻性黄疸大鼠肝脏功能及纤维化损伤的保护作用。方法 32只SD大鼠随机分为对照组、梗黄组、高剂量干预组及低剂量干预组,梗黄组、高剂量组及低剂量组采用胆总管结扎法建立梗阻性黄疸大鼠模型,高剂量组[20μg/(kg.d)]及低剂量组[10μg/(kg.d)]采用外源性甲状腺素进行干预,4周后检测各组大鼠血清甲状腺激素水平、肝组织氧化还原酶系活力及肝纤维化指标。结果造模4周后,高剂量组、低剂量组、梗黄组血清促甲状腺激素(thyroid stimulating hormone,TSH)水平低于对照组(均P<0.05),高剂量组、低剂量组及对照组血清游离甲状腺素(FT3、FT4)水平高于梗黄组(均P<0.05)。高剂量组、低剂量组肝组织匀浆超氧化物歧化酶(superoxygen dehydrogenises,SOD)水平高于梗黄组(均P<0.05),丙二醛(methane dicarboxylic aldehyde,MDA)、层粘连蛋白(laminin,LA)、透明质酸酶(hyaluronidase,HA)、Ⅲ型前胶原(typeⅢprocollagen,PC-Ⅲ)及Ⅳ型胶原(typeⅣcolla-gen,Ⅳ-C)水平低于梗黄组(均P<0.05)。结论外源性甲状腺素能够改善梗阻性黄疸大鼠甲状腺激素代谢异常,减轻肝脏的氧化损伤,改善梗阻性黄疸大鼠肝脏纤维化。

【Abstract】 Objective To investigate the protect effects of ecyogentic thyroxine for hepatic fibrosis on rats with obstructive jaundice.Methods 32 SD rats were randomly divided into control group(CG),obstructive jaundice group(OG),high-dose group [HG,20 μg/(kg·d)] and low-dose group [LG,10 μg/(kg·d)],obstructive jaundice model were established with ligating-mutilation bile duct in OG,HG and LG,ecyogentic thyroxine were taken in HG and LG,after 4 weeks,levels of thyroxinein serum,activity of oxidoreduction enzyme system and levels of hepatic fibrosis index were detected in all rats.Results At 4 weeks after establishing medol,levels of thyroid stimulating hormone(TSH) in serum were lower in OG,HG and LG than in CG(all P < 0.05),levels of FT3 and FT4 in serum were higher in CG,HG and LG than in OG(all P < 0.05),levels of SOD in hepatic tissue homogenate were higher in HG and LG than in OG(all P < 0.05),levels of MDA,LA,HA,PC-Ⅲ and Ⅳ-C in hepatic tissue homogenate were lower in HG and LG than in OG(all P < 0.05).Conclusion Ecyogentic thyroxine can improve abnormality of thyroxine metabolism,lessen injury of oxidative stress and improve hepatic fibrosis on rats with obstructive jaundice.
