去卵巢大鼠行为改变与海马结构中突触素表达减少的相关性研究
Correlatation study of behavior changes of the ovariectomized rats and the reduction of synaptophysin expression in hippocampal formation
【摘要】 目的通过去卵巢大鼠的模型,观察雌激素缺乏后对学习和记忆的影响以及突触素表达的情况。方法实验选用成年Wistar大鼠33只,随机分为对照组、去卵巢组、补充雌激素组(去卵巢),每组11只,实验周期为4个月,观察指标:血中的雌二醇含量、记忆跳台潜伏期和错误次数以及海马结构中突触素表达情况。结果①血清中雌二醇浓度:对照组为(46.7±25.3)pg/mL;去卵巢组为(0.3±0.3)pg/mL;补充雌激素组为(144.4±78.9)pg/mL,各组间比较差异均有统计学意义(P<0.05)。②跳台记忆潜伏期和错误次数:对照组分别为(148.7±19.6)s、(2.9±1.1)次;去卵巢组为(85.6±16.8)s、(7.9±2.9)次;补充雌激素组为(101.6±18.6)s、(4.6±1.1)次;除了去卵巢组的记忆潜伏期与补充雌激素组比较无显著差异外(P>0.05),其他各组之间差异均有统计学意义(P<0.05)。③海马结构突触素的表达:去卵巢组海马的神经元胞浆内有少许棕黄色的颗粒,呈弱阳性;补充雌激素组与对照组相似,呈强阳性表达。结论雌激素缺乏可以导致大鼠跳台试验的错误次数增加和记忆潜伏期延长,突触素表达减少;补充雌激素有所好转,同时突触素表达增多,推测大鼠雌激素缺乏后的认知功能减退可能与海马结构中突触素表达减少有关。
【Abstract】 Objective To observe the effects of estrogen deficiency on the abilities of learning and memory as well as the synaptophysin expression,by using the ovariectomized rats’ models.Methods 33 Wistar rats were randomly divided into 3 groups: control group,ovariectomized(OVX) group and OVX+E2 group(ovariectomy and estrogen replacement therapy group),each group had 11 rats,the experimental period was 4 months,the indicators of estradiol concentration in serum,memory latency and error numbers in step-down test were observed,as well as synaptophysin expression in the hippocampal formation(HF).Results ①serum estradiol concentrations: control group was(46.7±25.3)pg/mL;OVX group was(0.3±0.3)pg/mL;OVX+E2 group was(144.4±78.9)pg/mL;significant differences were observed among the three groups.② memory latency and error numbers in step-down test: control group was separately(148.7±19.6) s,(2.9±1.1) times;OVX group was separately(85.6±16.8) s,(7.9±2.9) times;OVX+E2 group was separately(101.6±18.6) s,(4.6±1.1) times;no significant difference was found except memory latency in OVX group comparing with the value in OVX+E2 group,there were significant differences between other each in two groups.③synaptophysin expression in HF: a little brown-yellow granules within the cytoplasm of neuron were observed,which expression was weakly positive;the expressions in OVX group was intensely positive,which similar as the ones in control group.Conclusion Estrogen deficiency results in increase of the error numbers and elongation of the memory latency in step-down test,meanwhile synaptophysin expression decreases;it takes a favorable turn after replenishing estrogen and synaptophysin expression increases,so the cognitive function impairment after estrogen deficiency maybe relate to the reduction of synaptophysin expression in the HF.
