【摘要】 目的观察芹菜素对人结肠癌细胞SW1116及CIK细胞的作用,探讨芹菜素对人CIK细胞杀伤结肠癌SW1116细胞的影响。方法不同浓度的芹菜素分别诱导人CIK细胞和结肠癌细胞SW1116细胞24、48、72 h,四甲基偶唑蓝(MTT)法检测上述两种细胞的生长。乳酸脱氢酶释放法检测CIK细胞对结肠癌SW1116细胞杀伤活性。结果体外培养10 d后,CIK细胞比例由3.12%增加到17.55%。与对照组比较,浓度1.17～4.7μmol/L芹菜素作用后的CIK细胞的增殖率显著提高(P<0.05),浓度37.5～600μmol/L的芹菜素对SW1116细胞生长抑制率显著提高(P<0.05),三个时间段比较均无明显差异。浓度2.35～9.4μmol/L的芹菜素作用48 h后,CIK细胞对结肠癌SW1116细胞的杀伤活性显著高于对照组(P<0.05)。结论芹菜素在一定浓度范围内可促进CIK细胞的生长,明显提高CIK细胞对SW1116细胞的杀伤作用,且抑制结肠癌SW1116细胞的生长。更多还原

【Abstract】 Objective To observe the effect of apigenin on colonic cancer SW1116 cells and CIK cells,and explore its influence on CIK cells killing colonic cancer SW1116 cell.Methods CIK cells and colonic cancer cell SW1116 were treated with different concentrations of apigenin for 24 h,48 h,72 h,the cell proliferations were measured by methyl thiazolyl tetrazolium(MTT).The cell-killing activity of CIK cells to SW-1116 cells were detected by lactate dehydrogenase release.Results After 10 days in vitro culture,CIK cells increased from 3.12% to 17.55%.Compared to the control group,apigenin at the concentration of 1.17-4.7 μmol/L,could proliferate the CIK cell growth significantly(P<0.05),apigenin at the concentration of 37.5-600μmol/L could inhibit colonic cancer cells lines SW1116 significantly(P<0.05),no obvious difference observed between the three time periods;after 48 h apigenin culture at the concentration of 2.35-9.4 μmol/L the killing activity of CIK cells to the SW1116 cells increased significantly(P<0.05).Conclusion Apigenin with given concentration can promote the growth of CIK cells and improve the killing activity of CIK to SW1116 cells,and suppress the growth of colonic cancer cell SW1116.更多还原
