TM和EPCR基因在百草枯中毒鼠肺组织中的表达及三七总皂甙保护作用

【摘要】 目的探讨急性百草枯(PQ)中毒鼠肺组织病理损伤变化和肺组织中血栓调节蛋白(TM)和内皮细胞蛋白C受体(EPCR)的表达及三七总皂甙(PNS)的保护作用。方法 150只SD雄性鼠分为正常对照组(C 30只)、百草枯染毒组(PQ 60只)及三七总皂甙干预组(PNS 60只)。PQ组和PNS组鼠一次性灌胃PQ25mg/kg染毒,C组予等体积生理盐水灌胃。其中PNS组鼠于染毒前15min以PNS50mg/kg阴茎静脉注射保护,以后每日1次给药直至处死前;PQ组、C组分别在同时间点予与等体积生理盐水。观察各组大鼠在中毒后6、12h、1、3、5、7天肺组织病理改变,采用反转录-聚合酶链反应(RT-PCR)方法测定鼠肺组织TM和EPCR mRNA的表达。结果 C组TM和EPCR mRNA有一定水平表达;与C组相比PQ组及PNS组鼠肺组织TM和EPCRmRNA的表达增强,差异有统计学意义(P<0.05);PQ组TM和EPCR mRNA的表达在1天达高峰之后下降,第3天基本恢复到C组水平;PNS组TM和EPCR mRNA的表达在6、12h及1天低于PQ组,至第3和5天显著高于PQ组,之后下降至正常水平,差异有统计学意义(P<0.05)。PQ组肺病理损伤评分在6、12h,1、3、5及7天各亚组均高于PNS相应组,差异有统计学意义(P<0.05),C组肺组织病理大致正常,与PQ组及PNS组相比,差异有统计学意义(P<0.00)。结论 TM和EPCR mRNA基因参与PQ中毒所致肺损伤,PNS对百草枯中毒所致肺损伤有保护作用。

【Abstract】 Objective To explore the expressions of thrombomodulin(TM) and endothelial protein C receptor(EPRC) in lung of paraquat poisoning rats and the protection of Panax Notoginseng Saponins(PNS).Methods One hundred and fifty SD male rats were randomly divided into 3 groups:normal control group(C,30rats),paraquat poisoning group(PQ,70rats) and drug intervention group(PNS,70rats).The PQ and PNS group rats were given paraquat by gastric perfusion(25mg/kg).The C group rats were given saline by gastric perfusion.The rats in PNS group were protected by intravenous injections of penis with PNS(50mg/kg)15min before exposure of paraquat,once a day until killed.Ratsin the PQ and C group were given saline.Histological changes of lung tissues under light microscope were observed.And the expression of TM and EPCR mRNA in lung tissues of rats were measured by RT-PCR at different time of 6h,12h,1day,3days and 7days after poisoning.Results The relative magnitude expression of TM and EPCR mRNA in PQ(6h),PQ(12h)and PQ(1day)subgroups were significantly higher as compared with the PNS group(P<0.05).PNS(3d) and PNS(1day) subgroups were significantly higher as compared with PQ group.The pathological scores of lung injury in PQ(6h),PQ(12h),PQ(1day)and PQ(3days)subgroups were significantly higher compared with PNS group.Conclusion The aggravation of lung injury induced by paraquat in mice was related to the expression of TM and EPCR mRNA in lung tissue.PNS can effectively improve the expression of TM and EPCR mRNA in lung tissue and reduce the pathological changes of lung tissue.
