【摘要】 目的观察不同浓度葛仙汤对实验性溃疡性结肠炎(UC)大鼠结肠组织肿瘤坏死因子(TNF-α)和诱生型一氧化氮合酶(iNOS)蛋白表达的影响,探讨葛仙汤改善UC症状的可能机制。方法随机将SD大鼠分为4组,包括模型对照组和葛仙汤高、中、低剂量治疗组,采用大肠杆菌加乙酸复合法建立溃疡性结肠炎大鼠模型,连续给药21天后处死,进行结肠组织病理学评分(histopathological score,HS),免疫组化法检测各组大鼠结肠组织中TNF-α和iNOS蛋白的表达。结果用药3周后,低、中、高剂量葛仙汤治疗组的结肠HS均明显低于模型对照组(P<0.01);3个治疗组的结肠组织中TNF-α和iNOS蛋白阳性表达积分值较模型对照组明显降低(P值<0.01)。结论葛仙汤能明显减轻溃疡性结肠炎大鼠结肠的组织学损伤程度,可能与其下调TNF-α和iNOS蛋白的表达有关,其低剂量即发挥疗效。更多还原

【Abstract】 Objective To observe the impact of Gexiantang(GXT) in different concentration on TNF-α and iNOS expression in colonic tissue of rats with experimental ulcerative colitis(UC) and to investigate the possible mechanism of Gexiantang on the improvement of common symptoms for UC.Methods We randomly assigned the SD rats into four groups,including the control group,high dosage GXT group,medium dosage GXT group and low dosage GXT group.We used combined escherichia coli and acetate acid method to establish UC rats model.After 21 days treatment,all rats were sacrificed,and their colonic tissue were measured by histopathological score(HS).Expression of TNF-αand iNOS were detected by immunohistochemistry in each group.Results After 3 weeks treatment,compared with the control group,the colonic HS in the 3 GXT treatment groups were lower(P<0.01).Expression of positive integral value TNF-αand iNOS of the colonic tissue in the treatment group showed significant lower(P<0.01) than that in control group.Conclusion These results suggest that GXT demonstrates significant result on reducing histological damage of colonic tissue for UC rats.It is possible related with declined expression of TNF-α and iNOS.GXT in low dosage has showed clinical efficacy.更多还原
