CACNB2基因多态性与钙离子通道阻滞剂降压疗效的相关性研究

【摘要】 目的探讨钙离子通道辅助β亚基(CACNB2)6个SNP位点多态性对单一钙离子通道阻滞剂(calcium channelblockers,CCB)治疗原发性高血压疗效的影响。方法 104例原发性高血压患者同时服用单一钙离子通道阻滞剂(纳欣同)20mg,每日1次,随访6周,采用多重聚合酶链反应(multi-PCR)方法及基质辅助激光解析-飞行时间质谱分析技术(MLD1-TOFMS)对患者CACNB2的6个SNP进行基因分型检测,研究不同SNP位点以及同一SNP不同分型对CCB治疗高血压的疗效差异。结果 rs11014166 AA基因型患者服用钙拮抗剂的降压有效率高于AT+TT基因型患者,差异有统计学意义(P<0.05)。其余5个SNP位点不同基因型患者用钙拮抗剂的降压效果差异无统计学意义(P>0.05)。结论 CACNB2 rs11014166多态性与钙拮抗剂降压疗效相关,AA基因型人群对钙拮抗剂较为敏感。

【Abstract】 Objective To investigate the association between 6 single nucleotide polymorphisms(SNP) of L-calcium channel auxiliary subunit β2 gene(CACNB2) and the efficacy of calcium channel blockers(CCB) in the treatment of essential hypertension.Methods A total of 104 EH patients were given 20mg CCB(extended release nifedipine tablets) monotherapy everyday for six weeks.Six SNPs in the CACNB2 determined by multiplex polymerase chain reaction(multi-PCR) and matrix assisted laser desorption ionization time of flight MS(MLD1-TOF MS) to analyze the association of the CCB-efficacy for hypertension with different SNPs and their genotypes.Results The AA genotype of rs11014166 was more efficient than AT and AA genotype(P<0.05).Efficacy of genotypes of other five SNPs was not different each other(P>0.05).Conclusion The rs11014166 polymorphism of CACNB2 was associated with curative effect of L-calcium channel blockers,which was more effective in patients with AA genotype EH.
