【摘要】 目的通过检测安徽汉族人中急性心肌梗死(AMI)患者及健康对照者的血小板膜糖蛋白(GP)受体Ⅰa C807T基因多态性,明确血小板GPⅠa C807T的基因型,探讨其多态性与AMI之间的关系。方法提取人基因组DNA,用多聚酶链反应(PCR)扩增AMI组(74例)和健康对照组(60例)外周血DNA的基因片段,用琼脂糖凝胶分离扩增产物,在紫外透射仪下观察结果并分析。结果血小板膜糖蛋白Ⅰa-807基因多态性分析:AMI组的GPⅠa807TT+TC基因型携带者频率显著高于对照组。结论Ⅰa 807TT+TC的高表达是AMI患者的重要危险指标,Ⅰa C807T多态性可能与AMI的发生密切相关。更多还原

【Abstract】 Objective To detect the platelet glycoprotein(GP) receptor Ⅰa C807T gene polymorphism in Chinese Han population,and to investigate the possible relation ship between acute myocardial infarction(AMI) and platelet GP Ⅰa C807T gene polymorphism.Methods In 74 patients with AMI and 60 control subjects,the GP Ⅰa gene 807nt polymorphisms were detected by polymerase chain reaction amplication of fragments of the genomic DNA by using specific oligonucleotide primers in a thermal cycler.Restricted DNA products were then separated by agarose gel electrophoresis and visuslized by UV light.Results The frequence of GPⅠa gene 807nt TT+TC genotypes in the AMI group were higher than that in the control group.Conclusion The results suggest that there is a strong association between the presence of GP Ia T allele and AMI.GP Ia-807 T genotype is a major risk factor of acute myocardial infarction.更多还原
