烟草花叶病毒快速检测方法的建立及应用

【摘要】 应用单克隆抗体技术获得抗烟草花叶病毒(Tobacco mosaic virus,TMV)单克隆抗体,并通过胶体金标记技术,免疫层析快速检测技术和SQUID磁学定量测定技术建立起烟草花叶病毒的快速检测方法,并开发出用于定性检测的TMV胶体金快速检测试纸及用于定量检测的TMV纳米磁珠快速检测试纸,其检测灵敏度分别为1 ng/mL和0.1 ng/mL.所建立的方法操作简单,快速,灵敏度高,特异性好,是作为烟草育苗中各种TMV无症和有症烟株现场筛查的有效手段.

【Abstract】 The anti-TMV monoclonal antibodies were obtained through monoclonal antibody technology.The rapid test method of tobacco mosaic virus was established through the integration of colloidal gold labeling,immunoassay and quantitative determination of SQUID magnetic technology.The colloidal gold test strips were established for qualitative detection of TMV and the detection sensitivity was 1ng/mL.The magnetic immunoassay strips were established for quantitative detection of TMV and the detection sensitivity was 0.1ng/mL.Both methods were simple,rapid,high sensitivity and specificity.It’s feasible to the rapid on-site screening of the TMV disease and disease-free tobacco plants for tobacco seedlings.
