覆盆子中山柰酚的TLC鉴别和HPLC测定

【摘要】 目的:研究覆盆子药材中山柰酚的TLC鉴别和HPLC测定方法。方法:以硅胶G为固定相,以甲苯-乙酸乙酯-甲酸(9∶3∶1)为展开剂,1%三氯化铝乙醇液为显色剂,建立覆盆子药材中山柰酚的TLC鉴别方法;以甲醇-0.4%磷酸(55∶45)为流动相,大连依利特色谱柱,流速1mL·min-1,在360nm处进行山柰酚的HPLC测定。结果:TLC能检测出山柰酚的特征斑点;山柰酚在HPLC进样0.0104~0.052μg线性关系良好。结论:该方法简单、可行,可用于覆盆子药材的质量控制。

【Abstract】 Objective: To study the identification method by TLC and the determination method by HPLC of kaempferol in Rubi Fructus. Methods: TLC was used in the identification, silica G was used as the stationary phase, the proportion of methylbenzene-ethyl acetate-methanoic acid(9∶3∶1) was the mobile phase. The plates were sprayed with 1% aluminium choride of ethanol and heated to 105 ℃ for 5 min. HPLC was used in the determination. A C18 column was used with a mixture of methanol and 0.4% phosphoric acid solution(55∶45) as the mobile phase, the rate was 1 mL·min-1 with the detection wavelength at 360 nm. Results: The characteristic spot of kaempferol was examined by TLC, the method was proved to be linear over the range of 0.010 4~0.052 μg for kaempferol by HPLC. Conclusion: The method is simple and feasible, it provides a method for the quality control of Rubi Fructus.
