鞘内注射IL-1ra对CCI大鼠神经病理性疼痛的影响及可能机制
Effects and mechanisms of intrathecal injection of IL-1ra on neuropathic pain of chronic constrictive injury in rats
【摘要】 目的 观察白细胞介素1受体拮抗剂(interleukin-1 receptor antagonist,IL-1ra)对坐骨神经慢性压迫性损伤(chronic constriction injury,CCI)大鼠神经病理性疼痛的影响及可能机制。方法 采用大鼠CCI模型。雄性SD大鼠随机分为假手术组、CCI组、IL-1ra 10 mmg/L组和IL-1ra 1 mg/L组(n=8)。von Frey纤维丝和热痛刺激仪检测大鼠机械性缩足反射阈值和热缩足反射潜伏期,分别于术后第3、5、7天各时间点将大鼠处死,应用免疫组织化学检测脊髓背角神经元磷酸化细胞外信号调节激酶(p-ERK)表达的情况。结果 IL-1ra 10 mg/L可减轻CCI大鼠术后第3、5、7天机械痛敏与热痛敏,减少脊髓背角神经元中p-ERK表达,其中第5天尤为明显。结论 IL-1ra能减轻神经病理性疼痛,其机制可能与阻断IL-1β、降低p-ERK表达有关。
【Abstract】 Objective To investigate the effects and mechanisms of interleukin-1 receptor antagonist(IL-1ra)on neuropathic pain of chronic constrictive injury of the sciatic nerve in rats.Methods Male SD rats were randomly divided into 4 groups:sham operation group;control group(treated with CCI);IL-1ra 10 mg/L group;IL-1ra 1 mg/L group.Thermal withdrawal latency(TWL) and mechanical withdrawal threshold(MWT) of rats were determined by Hargreave’s thermal radiation apparatus and von Frey’s method of microfilament,respectively.The animals were killed at day 3,day 5 and day 7.Then expressions of phospho-extracellular signal-regulated kinase(p-ERK) were assessed by immunohistochemical analysis and Western blot method.Results IL-1ra could significantly alleviate thermal and mechanical hyperalgesia induced by CCI and attenuate the activation of p-ERK simultaneously at day 3,day 5 and day7.The effect of IL-1ra was the highest at day 5.Conclusion This study shows that IL-1ra can alleviate neuropathic pain,and its mechanism may be related to blocking IL-1β and reducing the expression of p-ERK.
