雷帕霉素对人胃癌SGC-7901细胞增殖的影响及其分子机制

【摘要】 目的 观察雷帕霉素对人胃癌SGC-7901细胞增殖的影响及其对缺氧诱导因子1α(HIF-1α)、血管内皮生长因子(VEGF)基因表达的影响。方法 SGC-7901细胞经雷帕霉素处理后,以四甲基偶氮唑蓝(MTT)法检测细胞的增殖情况,逆转录聚合酶链反应(RT-PCR)检HIF-1α、VEGF基因的表达。结果 体外雷帕霉素可抑制人胃癌SGC-7901细胞增殖,并下调人胃癌SGC-7901细胞HIF-1α、VEGF基因的表达。结论 雷帕霉素可抑制人胃癌SGC-7901细胞增殖,并且可能是通过下调人胃癌SGC-7901细胞HIF-1α、VEGF基因的表达而起到抑制作用的。

【Abstract】 Objective To investigate the effect of rapamycin on the proliferation of gastric cancer cell line SGC-7901,as well as hypoxia-inducible factor-1α(HIF-1α),vascular endothelial growth factor(VEGF) expression.Methods Gastric cancer cells were treated with rapamycin.Cell proliferation was detected by MTT assay.Semi-quantitative RT-PCR was applied to detect the expression of HIF-1α and VEGF mRNA changes.Results Rapamycin inhibited proliferation of human gastric cancer SGC-7901 cells in vitro.Rapamycin in human gastric cancer cells reduced HIF-1α,VEGF mRNA expression.Conclusion Rapamycin can inhibit proliferation of human gastric cancer SGC-7901 cells in vitro.Rapamycin may downregulate HIF-1α and VEGF mRNA expression in human gastric cancer cells.
