N-cadherin在小鼠黑质多巴胺能神经元慢性损伤过程中的表达变化
The expression of N-cadherin in substantia nigra of mice during the chronic damage of dopaminergic neurons
【摘要】 目的 探索神经型钙黏蛋白(N-cadherin)在多巴胺能神经元慢性损伤中的可能作用。方法 采用1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)腹腔注射建立小鼠黑质多巴胺能神经元慢性损伤模型,采用免疫荧光染色和Western blot方法检测小鼠从实验第1周到第6周黑质酪氨酸羟化酶(TH)和N-cadherin的表达情况。结果 免疫荧光染色结果显示:对照组和MPTP组小鼠黑质的TH免疫阳性神经细胞在各时间点都同时表达N-cadherin。Western blot结果显示:(1)MPTP组小鼠黑质的TH表达量从第3周开始呈梯度下降(P<0.05);而N-cadherin表达量从第4周开始突然下降(P<0.05),但在第4到6周内的表达量虽有波动但差异无统计学意义(P<0.05)。(2)虽然TH和N-cadherin的表达变化时程曲线似有区别,但相关分析显示两者之间仍存在正相关(r=0.924,P<0.05)。结论 本研究证明多巴胺能神经元损伤影响了胞内N-cadherin的表达,影响程度虽与神经元的损伤程度有关,但并不完全一致,其内在关系有待进一步研究。
【Abstract】 Objective To explore the possible roles of neural cadherin(N-cadherin) during the chronic damage of dopaminergic neurons.Methods Dopaminergic neurons chronic damage model was established by intraperitoneal injection of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyritine(MPTP).Immunofluorescence and Western blot were applied to detect the expression of tyrosine hydroxylase(TH) and N-cadherin in the mice substantia nigra(SN) from the first week to the sixth week.Results The immunofluorescence double-labeled results showed that N-cadherin positive cells were obviously present in mice SN at all of the time points and co-expressed with TH in the same neurons in control group and MPTP group.Western blot showed that:(1)TH protein began to decrease from the third week gradiently after MPTP injection(P<0.05).N-cadherin decreased from the forth week suddenly(P<0.05).(2)Although the expression of TH and N-cadherin seemed to be different,the correlation analysis of TH protein with N-cadherin showed that they were postitively correlated(r=0.924,P <0.05).Conclusion Our results suggest that the chronic damage of dopaminergic neurons affects the expression of N-cadherin.The influence is associated with the dopaminergic neurons damage but not exactly the same,which requires further study of their intrinsic relationship.
