Toll样受体2基因T597C多态性与中国南方汉族人群结核性脑膜炎的易感相关性

【摘要】 目的探讨Toll样受体2(TLR2)基因T597C单核苷酸多态性与中国南方汉族人群结核性脑膜炎的易感相关性。方法结核性脑膜炎组为2009年9月至2011年5月南方医院收治的确诊结核性脑膜炎患者9例,肺结核组为广州市胸科医院收治的肺结核患者20例,另选20例健康人作为对照组,各组均符合相应的纳入及排除标准。采用聚合酶链式反应及基因测序方法对TLR2基因T597C单核苷酸多态性与中国南方汉族人群结核性脑膜炎易感性进行相关分析。结果在结核性脑膜炎组中变异基因型(TC和CC)和597C等位基因的频率明显高于对照组,但变异基因型在对照组、肺结核组及结核性脑膜炎组间的频率分布差异无统计学意义(χ2=3.560,P=0.169),597C等位基因在各组间的频率分布差异无统计学意义(χ2=5.108,P=0.078)。结论尚不能证明TLR2基因T597C单核苷酸多态性与中国南方汉族人群结核性脑膜炎及肺结核的易感性相关。

【Abstract】 Objective To explore the associativity of T597C single nucleotide polymorphism(SNP) of Toll-like receptors 2(TLR2) gene with susceptibility to tuberculous meningitis(TBM) in the Han population of southern China. Methods From September 2009 to May 2011,nine Han patients with TBM were enrolled from Nanfang Hospital(TBM group),and twenty patients with pulmonary tuberculosis were selected from Guangzhou Chest Hospital(pulmonary tuberculosis group).Twenty health persons were used as the control group.The samples in each group were consistent with the inclusion criteria and exclusive criteria.Polymerase chain reaction and nucleotide sequencing were performed on DNA samples to identify allele and genotypes of T597C of TLR2,and the associativity of T597C polymorphisms of TLR2 gene with susceptibility to tuberculous meningitis was analysed. Results The frequency of the genotype variants(T/C and C/C) and TLR2 allele 597C in the TBM group was much higher than in the control group,but the frequency distribution of the genotype variants has no significant difference among the control group,pulmonary tuberculosis group and TBM group(χ2=3.560,P=0.169),neither the 597C allele(χ2=5.108,P=0.078). Conclusion The experiment can not confirm the association between the T597C SNP of TLR2 gene and the susceptibility to tuberculosis especially to TBM of Han population of southern China.
