白藜芦醇对视网膜母细胞瘤Y79细胞增殖的影响及其机制
Effects of resveratrol on proliferation of retinoblastoma Y79 cells and its mechanism
【摘要】 目的研究白藜芦醇(resveratrol,Res)对人视网膜母细胞瘤Y79细胞增殖、细胞周期和凋亡的影响及其机制。方法细胞计数法检测不同浓度Res处理Y79细胞后细胞的增殖活性,制作生长抑制曲线。流式细胞仪检测细胞周期和凋亡率,Western blot检测细胞NF-κB/p65和Survivin蛋白表达的变化。结果 Res能显著抑制Y79的增殖,其作用表现为呈剂量和时间依赖性(P<0.01)。流式细胞仪检测发现:100μmol.L-1Res处理Y79细胞后细胞阻滞于S期,从0h的(32.69±4.28)%上升至24h、48h、72h的(44.51±4.65)%、(55.41±3.14)%、(63.17±3.96)%;其凋亡率呈时间依赖性上升,从0h的(1.18±1.17)%上升至24h、48h、72h的(6.27±3.36)%、(11.13±4.04)%、(16.99±3.65)%,不同时间点间差异均有显著统计学意义(均为P<0.01)。Western blot检测发现:Y79细胞以100μmol.L-1Res处理后,NF-κB/p65表达从0h的0.53±0.09降至24h、48h、72h的0.44±0.10、0.37±0.10、0.24±0.09,Survivin表达从0h的0.73±0.12降至24h、48h、72h的0.62±0.11、0.43±0.11、0.24±0.09,不同时间点间差异有统计学意义(P<0.05或0.01)。结论 Res对人视网膜母细胞瘤Y79细胞具有明显的增殖抑制作用,其作用机制与细胞周期阻滞和诱导凋亡有关。
【Abstract】 Objective To study the effects of resveratrol(Res) on proliferation,cell cycle and apoptosis of retinoblastoma Y79 cell and its mechanism.Methods Cell counting method was used to observe the proliferation of Y79 cells deal with different concentrations of Res,and the growth curve was mapped.Flow cytometry was used to measure the expression of cell cycle and apoptotic rate.Western blot was used to measure the expression of NF-κB/p65 and Survivin.Results Res obviously inhibited growth of Y79 cells in a dose-dependent and time-dependent manner(P<0.01).Flow cytometry showed the Y79 cell stayed in S stage after treated with 100 μmol·L-1 Res,cell in S stage increased from 0 hour(32.69±4.28)% to 24 hours(44.51±4.65)%,48 hours(55.41±3.14)%,72 hours(63.17±3.96)%,and apoptosis rate increased in a time-dependent manner,which were from 0 hour(1.18±1.17)% to 24 hours(6.27±3.36)%,48 hours(11.13±4.04)% and 72 hours(16.99±3.65)%,there were significant differences between each two time points(all P<0.01).Western blot showed that,after treated with 100 μmol·L-1 Res,NF-κB/p65 expression decreased from 0 hour(0.53±0.09) to 24 hours(0.44±0.10),48 hours(0.37±0.10),72 hours(0.24±0.09),and Survivin expression decreased from 0 hour(0.73±0.12) to 24 hours(0.62±0.11),48 hours(0.43±0.11) and 72 hours(0.24±0.09),there were statistical difference between each two time points(P<0.05 or 0.01).Conclusion Res can obviously inhibit the proliferation of human retinoblastoma Y79 cells,and its mechanism is related to blocked cell cycle and induced apoptosis.
