野扁桃AlsCBF基因植物表达载体的构建
Structure of Plant Expression Vectors of AlsCBF Gene from Amygdalus ledebouriana Schlecht
【摘要】 以AlsCBF基因cDNA和植物表达载体pCAMBIA1301进行双酶切,将抗寒基因AlsCBF与表达载体正向选择系统结合,构建抗寒基因植物表达载体pCA1301-AlsCBF。对阳性克隆通过菌落PCR、酶切鉴定和测序分析,成功将目的基因序列正确连接到pCAMBIA1301,构建其植物表达载体,并转入根癌农杆菌LBA4404中。
【Abstract】 This study was carried out to cut double enzyme with AlsCBF gene cDNA and plant expression vectors pCAMBIA1301,to combine cold-resistant gene AlsCBF with positive selection system to construct plant expression vectors of cold-resistant gene.In positive cloning,through the colonies PCR,enzymatic digestion and sequencing analysis,the purpose gene sequence will be properly connected with the pCAMBIA1301 to build the plant expression vectors and to be moved on into Agrobacterium tumefaciens LBA4404.
