不同质量浓度肉苁蓉总苷诱导大鼠骨髓间充质干细胞向神经细胞样细胞的定向分化

【摘要】 背景:肉苁蓉及其所含的化学成分有神经保护作用,并能促进神经功能的恢复。目的:分析肉苁蓉总苷体外诱导骨髓间充质干细胞分化为神经细胞样细胞的最佳剂量。方法:分离培养Wistar大鼠骨髓间充质干细胞,并利用β-巯基乙醇及不同质量浓度肉苁蓉总苷诱导骨髓间充质干细胞,分为空白对照组、β-巯基乙醇对照组、肉苁蓉总苷100,200,300μg/L组进行观察。结果与结论:诱导7d后,与空白对照组比较,其他各组活细胞数明显增高(P<0.05);诱导第3天,与空白对照组比较,内苁蓉总苷各剂量组细胞Nestin、NSE阳性表达率明显增高(P<0.05)。100,200μg/L质量浓度肉苁蓉总苷可有效促进骨髓间充质干细胞增殖,并能够明显促进其向神经细胞样细胞分化,300μg/L质量浓度可以促进骨髓间充质干细胞增殖,但效果不及100,200μg/L组(P<0.05)。

【Abstract】 BACKGROUND: Cistanche and its chemical composition play a neuroprotective role, and can promote recovery of neurological function. OBJECTIVE: To analyze the optimal dose of cistanche total glycosides for in vitro induction of bone marrow mesenchymal stem cells differentiating into nerve cells. METHODS: Bone marrow mesenchymal stem cells of Wistar rats were isolated and then cultured. β-mercaptoethanol and different concentrations of cistanche total glycosides were used to induce bone marrow mesenchymal stem cells. Bone marrow mesenchymal stem cells of Wistar rats were divided into blank control group, β-mercaptoethanol control group, cistanche total glycosides groups of 100, 200, 300 μg/L for observation. RESULTS AND CONCLUSION: After 7 days of induction, the number of living cells increased obviously in other groups, compared with the blank control group (P < 0.05); at the 3rd day of induction, Nestin, NSE-positive expression rate was significantly higher in cistanche total glycosides groups, compared with the blank control group (P < 0.05). 100, 200 μg/L cistanche total glycosides could effectively promote proliferation of bone marrow mesenchymal stem cells, and significantly promote the differentiation into nerve cells. 300 μg/L cistanche total glycosides could promote proliferation of bone marrow mesenchymal stem cells, but the effect was less than 100, 200 μg/L (P < 0.05).
