1株好氧堆肥高温H2S脱臭菌的筛选、鉴定及生长特性
Isolation,Identification and Growth Characteristics of a Thermophilic Bacterium for H2S Deodorization during Aerobic Composting
【摘要】 为减少好氧堆肥过程中硫化氢的释放,通过选择培养基进行富集培养筛选菌株,筛选能转化硫化氢的高温脱臭菌。结果从堆肥中筛选到1株高温细菌HB3,经生理生化特征分析、16S rDNA基因序列比对,鉴定该菌株为假黄色单胞菌属。该菌株在50℃条件下可将S2-形式的H2S转化为硫酸根形式,在硫化物为唯一硫源且其质量浓度为50 mg/L时,转化率最高可达36.89%;该菌株适宜在酸性条件下生长,45~55℃生长状态稳定,以硫化钠为底物生长速度最快。同时测定其在不同温度、pH和底物条件下的生长量。菌株HB3可在高温条件下生长并有较高的硫化氢转化率,具有较好的应用前景。
【Abstract】 In order to reduce the release of hydrogen sulfide during aerobic composting process,a thermophilic deodorizing bacterium strain capable to transform hydrogen sulfide was screened through selective medium to carry out enrichment culture.As the results a bacterium strain HB3 was screened from the composts.Analysis of its physio-biochemical characteristics,16S rDNA gene sequencing comparison,the strain was identified,and belonged to the genus of Pseudoxanthomonas.The bacterium could metabolize S2-of H2S into the form of SO42-under 50 ℃;Furthermore,when 50 mg/L of sodium sulfide was used as the sole sulfur source,conversion rate reached at the highest of 36.89%,and the strain had the aptitude to grow in acid condition,and stably grew at 45 to 55 ℃;and it grew the fastest when taking sodium sulfide as a substrate.The growth under conditions of different temperature,pH and substrate.The strain could grow and had fairly high conversion rate at high temperature,and had fairly good application prospect.
