S100A4和缺氧诱导因子-1α在胃癌中的表达及意义

【摘要】 目的探讨胃癌中S100A4和缺氧诱导因子-1α(HIF-1α)的表达及其与临床病理因素之间的关系。方法采用RT-PCR法和免疫组化SP法检测45例胃癌新鲜标本及其切端正常黏膜组织中S100A4和HIF-1α的表达情况。结果 RT-PCR结果示S100A4和HIF-1αmRNA在胃癌组织表达而在切端正常黏膜组织不表达,HIF-1α和S100A4 mRNA的表达与肿瘤淋巴结转移及临床病理分期有关(P<0.05,P<0.01);免疫组化结果示S00A4和HIF-1α蛋白在胃癌组织中的表达阳性率分别为48.9%和60%,而在切端正常黏膜组织不表达或表达不明显,HIF-1α和S100A4蛋白表达与肿瘤淋巴结转移、浸润深度和临床病理分期有关(P均<0.05)。S100A4和HIF-1α的表达呈显著正相关。结论 S100A4和HIF-1α的表达促进胃癌的发生发展;S100A4与HIF-1α间存在协同作用。

【Abstract】 Objective: To evaluate the expression and clinical significance of S100A4 and hypoxia-inducible factor-1α(HIF-1α) in gastric cancer.Methods: RT-PCR and immunohistochemistry were employed to examine the expression of S100A4 and HIF-1α in 45 cases of freshly gastric cancer tissue and normal gastric mucosa.Results:The results of RT-PCR revealed that there were S100A4 and HIF-1α mRNA expression in gastric cancer while S100A4 and HIF-1α mRNA were not found in any normal gastric mucosa.The expression of S100A4 and HIF-1α mRNA was closely related to lymph node metastasis and clinical pathological stage(P<0.05,P<0.01.The results of immunohistochemistry revealed that the expressions of S100A4 and HIF-1α proteins in gastric cancer were 48.9% and 60% respectively.The expressions of S100A4 and HIF-1α proteins were closely related to lymph node metastasis,depth of tumor invasion and clinical pathological stage(P<0.05).The expression of S100A4 had positive correlation with HIF-1α.Conclusion: S100A4 and HIF-1α may be involved in the pathogenesis of gastric cancer,S100A4 and HIF-1αmay have synergetic effects.
