酶解辅助提取关黄柏生物碱的工艺研究

【摘要】 目的:考察酶解辅助提取技术对关黄柏生物碱提取率的影响,优选出总生物碱的最佳提取工艺条件。方法:采用正交实验,考察酶解辅助法在不同pH,不同温度,不同浸取时间对关黄柏中总生物碱提取率的影响。结果:优化酶解法的最佳提取工艺条件为:纤维素酶用量为0.9%,在pH4.5,35℃条件下酶解2.5 h。酶解法可将总生物碱提取率提高约27%。结论:酶浸-回流法提取总生物碱方法可行,该方法提取时间短,提取率高,适合工业化生产。

【Abstract】 Objective: To inspect the effect of enzymolysis-assisted extraction technology of alkaloid in Phellodendron amurense,and optimize the best extraction technology of total alkaloid.Methods: Orthogonal test was designed for optimizing the enzymatic-assisted extraction of total alkaloid in Phellodendron amurense.The effect of enzyme dosage,pH,treatment temperature,treatment time to the extraction of total alkaloid in Phellodendron amurense were observed.Results: The optimum cellulose degradation were obtained as follows:The cellulose dosage by 0.9%,pH4.5,the reaction time of 2.5h and the reaction temperature of 35℃,By this method,extraction ratio could be improved 27%.Conclusion: The method of Enzymatic-assisted and Refluence-Cellulose extraction is workable,which should be shorted extraction time and increased extraction rate of total alkaloid.It is suitable for industrialized manufacture.
