EGCG对雨蛙素诱导的小鼠急性胰腺炎的治疗作用

【摘要】 目的:探讨EGCG对雨蛙素诱导的小鼠急性胰腺炎(acute pancreatitisAP)的治疗作用。方法:balb/c小鼠54只,随机分为对照组、模型组、治疗组,各组又分为造模后8、12、24h(各6只)。模型组采用腹腔注射雨蛙素(50μg/kg),7次,建立小鼠AP型。治疗组在造模后1、3、6h腹腔注射EGCG(25mg/kg)。造模成功后8、12、24h处死小鼠,切取胰腺,测定血清中AMY、TNF-α、IL-6、MDA的含量,测定胰腺组织NF-κB水平及胰腺组织学观察及组织病理学评分。结果:小鼠血清淀粉酶、IL-6、TNF-α、MDA、胰腺组织NF-κB表达、胰腺组织病理学评分的结果,模型组相应时间点与对照组相比均显著增高(P<0.05),治疗组应时间点与模型组相比相均显著降低(P<0.05)。结论:EGCG可通过抑制NF-κB的活性下调TNF-a、IL6表达和提高对氧自由基清除,从而减轻实验室小鼠胰腺炎胰腺组织损伤及全身炎症反应。

【Abstract】 Objective To study the effect of epigallocatechin-3-gallate(EGCG) on the pancreatic injury in mice with acute pancreatitis(AP).Methods Fifty-four balb/c mice were randomly divided into 9 subgroups:Control group,AP induction group and treatment group with EGCG.Each group was divided into 8 h、12 h、 24 h subgroups after the induction of AP(n = 6 in each subgroup).In the AP induction group,cerulein(50 μg/kg body weight) in 0.9% NaCl was intraperitoneally injected into animals every one hour for 7 times.In the EGCG treatment group,EGCG(25 mg/kg) was injected in abdominally 1,3,and 6 hours after the induction of pancreatitis.Six mice from each subgroup were killed for evaluation 8、12、24 h after the pancreatitis induction.The levels of AMS、 IL-6、 TNF-α、 MDA in serum and NF-κB activity were determined.HE staining was performed to evaluate the morphologic changes in animal pancreats.Results The level of AMS、 IL-6、 TNF-α、 MDA in serum,the pancreatic injury severity and NF-κB activity in pancreas in AP group were markedly higher than those in the control group(P < 0.05,respectively).In the EGCG treatment group,the levels of determination items decreased significantly than those in the AP group(P < 0.05).Conclusion EGCG could reduce pancreatic tissue injury in mice after AP treatment through inhibiting the NF-κB and oxidative stress pathways.
