小鼠骨髓间充质干细胞SCA-1~+/CD45~+/CD31~+亚群在心肌梗死中的作用
Roles of bone marrow mesenchymal stem cells subsets SCA-1~+/CD45~+/CD31~+ play in myocardial infarction in mice
【摘要】 目的:探讨小鼠骨髓间充质干细胞(mBMSCs)亚群SCA-1+/CD45+/CD31+在心肌修复中作用。方法:以小鼠心脏干细胞表面分化抗原检测mBMSCs后以CD45、CD31为标准分选得到四个亚群。各亚群及未分选群和心肌细胞共培养后检测α-actin、Cx43、Desmin、cTnI的表达。将SCA-1+/CD45+/CD31+注入心梗小鼠体内完成心功能测定及心脏DIR成像检查。结果:各组均表达心肌特异性分化抗原。SCA-1+/CD45+/CD31+表达的量更多。心脏彩超及心脏DIR成像提示SCA-1+/CD45+/CD31+改善心功能方面优于其他亚群及未分选群。结论:mBMSCs为一多克隆的细胞群体。SCA-1+/CD45+/CD31+向心肌定向分化及改善心功能方面优于其他亚群。
【Abstract】 Objective To investigate the roles of the SCA-1+/CD45+/CD31+ cells from bone marrow mesenchymal stem cells (BMSCs) in myocardium repair in mice. Methods The BMSCs were sorted by the differentiation antigens on the surface of mice heart stem cells, and then four subgroups were selected based on CD45 and CD31. The expressions of α-actin, Cx43, Desmin, cTnI were detected. The SCA-1+/CD45+/CD31+cells were injected into mice with myocardial infarction, then the cardiac color ultrasonography and heart DIR imaging were administered. Results The cardiac-specific differentiation antigen was expressed in each group, while the expression level was even higher in SCA-1+/CD45+/CD31+ cells. Cardiac color ultrasonography and heart DIR imaging results indicated that SCA-1+/CD45+/CD31+ cells were prominent in improving heart function as compared with other subgroups and undifferentiated subgroup. Conclusion BMSCs were polyclonal cells, in which the SCA-1+/CD45+/CD31+ cells were more effective in directing the cardiac committed differentiation and improving cardiac function.
