口腔粘膜下纤维性变病变组织中差异miRNA的研究

【摘要】 目的构建口腔粘膜下纤维性变(OSF)病变组织中差异miRNA表达谱。方法选取1例临床上具有典型病理特征的OSF组织及其配对正常组织,以正常组织为对照,部分病变组织行Affymetrix miRNA芯片实验,部分组织用于芯片后的进一步验证,同时进行生物信息学分析,并与前期OSF病变组织的差异基因表达谱对接,初步确定差异miRNA的可能靶标。结果所选病变组织属于OSF病变中期,miRNA芯片显示差异1.5倍以上的miRNA共有19个,其中OSF病变组织中上调13个,下调6个,miRNA特异性实时定量PCR验证结果与芯片结果一致,生物信息学分析及与前期差异基因表达谱对接显示其中的miR-23b、miR-203及miR-200c等可能参与了OSF病变过程及其后的癌变。结论 miRNAs可能在OSF病变过程中起着重要作用,并可能参与OSF的癌变过程,其进一步研究将有助于阐明OSF的分子发病机制。

【Abstract】 Objective To identify the differential miRNA expression profile in the tissue of oral submucous fibrosis(OSF).Methods The miRNA expression profile of a specimen with typical pathological feature of OSF was compared with its paired normal mucosa by Affymetrix miRNA Array.The remain parts of the specimen were used to verify the results of miRNA Array by miRNA-specific real-time PCR.Meanwhile,online-bioinformatics were used and the miRNA Array was docked to previous cDNA Array to screen the potential targets of the differential miRNA.Results The specimen of OSF for miRNA Array belonged to stage Ⅱ.The miRNA Array revealed nineteen miRNAs with differential expression above 1.5 times,among which thirteen were up-regulated and six were down-regulated.The results of miRNA-specific real-time PCR were the same with those of miRNA Array.Bio-information analysis and docking with prophase differential gene expression profile suggested miR-23b,miR-203 and miR-200c might participate in the progress of OSF and in the subsequent carcinogenesis.Conclusions miRNAs may play important roles in the pathogenesis of OSF and subsequent carcinogenesis.Further research will elicit the molecular pathogenesis of OSF.
