FQ-PCR技术在儿童尿液巨细胞病毒检测的临床应用

【摘要】 目的探讨实时荧光定量聚合酶链式反应(FQ-PCR)方法检测尿液巨细胞病毒在儿童CMV感染诊断中的意义。方法分别应用FQ-PCR和ELISA法检测75例确诊或高度怀疑为CMV感染和25例对照儿童尿CMV-DNA和血清CMV-IgM,比较病毒DNA与血清抗体的平行性和敏感性。结果 75例确诊或疑诊CMV活动性感染患儿中,检测到CMV-DNA 65例;检测到CMV-IgM 54例。两种方法阳性符合率为77%,FQ-PCR和ELISA法阳性检出率分别为88%和72%,差异有统计学意义(P<0.05)。结论 FQ-PCR法检测尿CMV-DNA对诊断活动性CMV感染具有良好的应用价值。

【Abstract】 Objective To explore the significance of determining urine cytomegalovinus(CMV) DNA in diagnosis of cytomegalovinus infection in children by fluorescent quantitative polymerase chain reaction(FQ-PCR) assay.Methods CMV-DNA and anti-CMV IgM in the urine of 75 children with confirmed or suspected CMV infection and 25 controls were determined by FQ-PCR and ELISA.The parallelism and sensitivity of CMV-DNA and anti-CMV IgM were compared.Results Among the 75 children with confirmed or suspected active CMV infection,CMV-DNA was detected from 65 children and CMV-IgM was detected from 54 children.The positive coincidence rate of FQ-PCR and ELISA was 77%.The positive rates of the two methods were 88% and 72%,respectively,and the difference was statistically significant(P<0.05).Conclusions FQ-PCR can be widely used in detection of CMV-DNA for diagnosis of active CMV infection in children.
