复方虫草提取物对实验性肺纤维化大鼠的影响
Effects of Cordyceps sinenisis extract on bleomycin-induced pulmonary fibrosis in rats
【摘要】 目的探讨复方虫草提取物对实验性肺纤维化大鼠的影响及其相关机制。方法采取大鼠气管内注入博莱霉素的方法制备肺纤维化模型,分别在给予复方虫草提取物后第7、14、28天处死大鼠后测定肺系数、肺组织羟脯氨酸(HYP)含量、谷胱甘肽(GSH)的活力、总抗氧化能力(T-AOC)水平及α-平滑肌动蛋白(α-SMA)、转化生长因子βl(TGF-βl)蛋白和mRNA表达水平;取固定部位肺组织做病理组织学检查。以泼尼松作为阳性药对照观察。结果复方虫草提取物能明显改善纤维化病变,降低肺系数、肺组织HYP含量及TGF-β1 mRNA、TGF-β1、α-SMA蛋白表达水平,尤其第28天显著(P<0.01或<0.05),且能明显升高GSH活力、T-AOC水平(P<0.01或0.05)。结论复方虫草提取物可减轻肺泡炎症和肺纤维化的范围及程度,其作用机制可能与抗氧化作用和降低TGF-β1的表达进而抑制肺肌成纤维细胞的活化有关。
【Abstract】 Objective To study the effects and interventional mechanisms of Cordyceps sinenisis extract on bleomycin-induced pulmonary fibrosis in rats.Methods The pulmonary fibrosis model of rat was established by intratracheal administration of bleomycin.The pulmonary index,pathomorphology,GSH,T-AOC,HYP,α-SMA,TGF-βl protein and gene were assessed at the 7 d,14 d,28 d after drug administration.Lung samples collected were stained with hematoxylin-eosin(HE) staining and scored.BLM-prednisone group was positive control.Results Cordyceps sinenisis extract could alleviate BLM-induced pulmonary fibrosis in rats.The pulmonary index,the amount of HYP,the expression of α-SMA,TGF-βl and gene in lung tissue were significantly decreased in Cordyceps sinenisis extract group compared with those of control group(P<0.01 or P<0.05),While the activity of GSH and higher the level of T-AOC in lung tissue were enhanced in Cordyceps sinenisis extract group(P<0.01 or P<0.05).Conclusion Cordyceps sinenisis extract has an obvious relieving effect on rat pulmonary fibrosis,probably by the reinforcement of antioxidant systems,decreasing the expression of TGF-β1 and inhibiting the proliferation of lung myofibroblast.
