塞来昔布抑制肺癌增殖及血管生成的实验研究

【摘要】 目的探讨选择性环氧化酶-2(COX-2)抑制剂塞来昔布(西乐葆)对A549肺癌移植瘤的抑制作用及其对肿瘤组织血管生成的影响。方法建立裸鼠肺癌细胞株A549移植瘤模型,药物组给予含1250ppm塞来昔布的饮水,连续28d,计算抑瘤率。采用免疫组化法研究移植瘤血管内皮生长因子(VEGF),微血管密度(MVD)的表达。结果药物组平均瘤重(0.65±0.20)g,对照组平均瘤重(1.45±0.30)g,两组有显著性差异(P<0.01),塞来昔布抑瘤率为57.23%。药物组与对照组VEGF积分分别为(0.184±0.022)和(0.295±0.032)(P<0.01),MVD平均值分别为(18.80±3.67)和(34.50±5.20)(P<0.01)。结论塞来昔布对A549肺癌移植瘤有明显抑制作用,其作用机制之一与抑制肿瘤血管生成相关。

【Abstract】 Objective To investigate the effects of celecoxib on transplanted A549 lung cancer and angiogenesis.Methods The model of A549 lung adenocarcinoma cell line xenografted neoplasia in BALB/C nude mice was made,treatment group began receiving 1250ppm celecoxib in the H20 for 28 days,all mice were sacrificed on the 28th day after tumor implantation.The expression of vascular endothelial growth factor(VEGF),microvessel density(MVD)in xenografted tumor tissue were analyzed with immunohistochemical staining.Results The average weight of tumor nodules in treatment group was(0.65±0.20)g,while in the control group it was(1.45±0.30)g,there is statistical difference(P<0.01),the inhibitory rate of celecoxib was 57.23%;the VEGF staining score of tumor tissue in treatment group and control group were(0.184±0.022) and(0.295±0.032)(P<0.01),respectively,the average MVD in two groups were(18.80±3.67) and(34.50±5.20)(P<0.01),separately.Conclusion Celexocib has inhibitory effects on A549 xenografted neoplasm in nude mice,induction of angiogenesis might be its important mechanisms.
