肺腺癌胸水肿瘤细胞的生物学及形态学研究
Experimental study on the morphology and biological Characteristics of the tumor cells from malignant pleural effusion(MPE) of human lung adenocarcinoma
【摘要】 目的:模拟体内环境,对胸水中的肿瘤细胞进行生物学及形态学研究。方法:取病理确诊肺腺癌胸膜转移的胸水肿瘤细胞,以肺腺癌原发灶肿瘤细胞为对比,对其进行形态学观察、MTT法绘制生长曲线、计算细胞倍增时间,流式细胞仪测定细胞周期分布及细胞凋亡率。结果:胸水肿瘤细胞生长曲线低平,倍增时间为(115.37±15.74)h,细胞周期分布G0+G1期(58.95±4.13)%、S期(26.2±3.7)%、G2期(14.85±1.3)%;细胞凋亡率,原发组及胸水组活细胞数占90%以上,原发灶肿瘤细胞悬浮培养48h后活细胞数仅为(38.9±6.86)%,扫描电镜示胸水肿瘤细胞膜微孔增大、纤毛呈球样及板样变。结论:肺腺癌原发灶肿瘤细胞与其胸水转移肿瘤细胞在生物学与形态学上存在差异。
【Abstract】 Objective:Simulating the environment of malignant pleural effusion(MPE),to conduct an investigation into the tumor cells from MPE of human lung adenocarcinoma on the morphology and biological characteristics.Methods:Compared with the cells from primary site,morphology and biological characteristics study were performed in the tumor cells from the MPE of human lung adenocarcinoma.The morphology of the tumor cells of the two groups(the primary site tumor cells and the MPE tumor cells) were observed with scan electron microscopy(SEM) and optical microscope,the cell viability was evaluated with Methyl thiazolyl tetrazolium(MTT) chromatometry assay and cell growth curve was generated.The tumor cells doubling time was calculated according to the cell growth curve,and the flow cytometry(FCM) was used to detect apoptosis percentage and phase distribution of cell cycles.Results:The growth curve of the tumor cells isolated from the MPE was a low and approximate straight line.Cell doubling time was(115.37 ± 15.74)h.Cell cycle distribution detection showed that cell cycle of the tumor cells from the MPE including :(58.95 ± 4.13)% cells in G0/G1 phase,(26.2 ± 3.7)% in S phase and(14.85 ± 1.3)% in G2/M phase.The number of viable cells in the primary site tumor group and MPE group were more than 90%.Suspended for 48 hours,the live rate of primary site tumor cell was only(38.9±6.86)%.Cell membrane of tumor cells from the MPE increased under SEM;cells cilia reduced,and cells were in a spherical sample and plate shape.Conclusion:There are significant differences between the tumor cells from MPE and the primary site tumor cells.
