β-连环蛋白在结肠癌细胞及肿瘤球细胞中的表达

【摘要】 目的:研究信号通路β-连环蛋白在人结肠癌ccl187细胞及其肿瘤球细胞中的表达状况。方法:培养ccl187细胞及其肿瘤球,分别提取ccl187细胞和肿瘤球细胞的总RNA,应用半定量RT-PCR和Westernblot方法分别检测β-连环蛋白在ccl187细胞及其肿瘤球中的mRNA水平及蛋白水平的表达。结果:半定量RT-PCR结果显示ccl187细胞和肿瘤球细胞中β-连环蛋白转录水平分别为41.6%和42.6%(P>0.05)。半定量Western blot结果显示ccl187细胞和肿瘤球细胞中β-连环蛋白的含量分别为36.7%和73.4%(P<0.05)。结论:信号通路β-连环蛋白在人结肠癌ccl187细胞及其肿瘤球细胞中转录水平基本一致,但肿瘤球细胞中β-连环蛋白被积累,含量显著高于ccl187细胞。

【Abstract】 Objective:To study the expression of signaling pathway β-catenin in human colon cancer ccl187cells and their tumor sphere cells.Methods:ccl187 cells and their tumor sphere cells were cultured.RNA was extracted respectively from ccl187 cells and their tumor sphere cells.The mRNA of β-catenin was examined by Semi-quantitive RT-PCR.The protein of β-catenin was examined by western blot.Results:Semi-quantitive RT-PCR results shown that the transcription level of ccl187 cells and their tumor sphere cells were 41.6% and 42.6% respectively(P>0.05).Western blot results showed that the translation level of ccl187 cells and their tumor sphere cells were 36.7% and 73.4% respectively(P<0.05).Conclusion:The transcription level of β-catenin of ccl187 cell and tumor sphere cells is comformable,but theβ-catenin protein is accumulated in tumor sphere cells.The amount of β-catenin protein in tumor sphere cells is higher than that of ccl187 cell markedly.
