银杏叶提取物对诱发性大鼠肝癌前病变的影响

【摘要】 目的:探讨银杏叶提取物对黄曲霉毒素B1(AFB1)诱发的大鼠肝癌前病变的影响。方法:71只大鼠随机分为3组:AFB1组、银杏叶提取物干预组、正常对照组。前2组予AFB1造模,银杏叶提取物干预组在造模的期间给予银杏叶提取物喂食大鼠。于实验第14、28、42及55周给予留取静脉血并予肝活检并于第64周全部处死大鼠,观察大鼠AFP的变化及肝脏γ-GT灶发生情况。结果:银杏叶提取物干预组大鼠血清AFP以及肝组织γ-GT灶的数量和大小均显著低于AFB1模型组(P=0.000)。结论:银杏叶提取物可减少AFB1诱发大鼠肝脏γ-GT灶的数量和大小以及延缓延缓血清AFP的升高,具有明显抑制AFB1诱发大鼠肝癌前病变的作用。

【Abstract】 Objective:To observe the effect of the extract of Ginkgo Biloba Leaf(EGb761) on aflatoxin B1(AFB1) induced precancerous lesion in Wistar rats.Methods:Seventy one wistar rats were randomized divided into three groups:Group AFB1,Group EGb761+AFB1 and Group of normal control.The experimental animals were injected with AFB1 through abdomen and the doses was(100-200)μg/kg bw,one to three times weekly in Group AFB1 and group EGb761+AFB1.The rats were taking in the food containing EGb761 in Group B only.Blood and biopsied liver samples were collected from all animals in each group at the experimental week of 14th,28th,42th and 55th.The animal experiment ended at 64th-week when the surviving animals were sacrificed by cervical dislocation.The serum specimens were stored at-40℃ for testing AFP by ELISA and AFB1-lysine adduct by HPLC.Gamma-glutamyl transpeptidase-positive hyperplastic cell foci(γ-GT foci) and histopathology of the liver tissue were observed.Results:The level of AFP(ng/ml) in the serum samples and the unmber and the size index of γ-GT foci of rat liver in group of EGb761+AFB1 were lower than that in group of AFB1(P=0.000).Conclusion:EGb761 is an effective inhibitor to precancerous lesion induced by AFB1 in rats.
