碱性成纤维细胞生长因子对人肝癌细胞蛋白激酶B活性的影响
Influence of basic fibroblast growth factor on protein kinase B activity in Bel-7402 cells
【摘要】 目的探讨碱性成纤维细胞生长因子(fibroblast growth factor,FGF)对人肝癌细胞(BEL-7402)蛋白激酶B(protein kinase B,PKB)活性的影响。方法以碱性FGF(b FGF)和wortmannin处理人肝癌细胞(BEL-7402),Western blot检测细胞外信号调节蛋白激酶B的活性。结果 25 ng/ml bFGF刺激人肝癌细胞(BEL-7402)10 min,细胞膜和胞质PKB酶活性达高峰。Wortmannin预处理后,人肝癌细胞(Bel-7402)细胞膜和胞质PKB活性在各时间点均明显降低。结论 bFGF能通过磷脂酰肌醇3-激酶(phosphatidylinositol 3-kinase,PI3K)途径迅速激活人肝癌细胞(BEL-7402)的PKB活性。
【Abstract】 Objective To study the influence of basic fibroblast growth factor(bFGF)on protein kinase B(PKB)activity in Bel-7402 cells.Methods Bel-7402 cells were treated with bFGF and wortmannin,and western blot was used to determine their PKB activities afterwards.Results PKB activity in both cell membrane and cytosol reached to the peak at 10 min after being treated by bFGF in a dose of 25 ng/ml.After Wortmannin pretreatment,PKB activities in both cell membrane and cytosol were significantly lower at each time point.Conclusions Basic fibroblast growth factor can rapidly activate PKB activity in both cell membrane and cytosol via P13K(phosphatidylinositol 3-kinase) in Bel-7402 cells.
