产低温脂肪酶深海菌株的筛选、鉴定及酶学性质研究

【摘要】 从鄂霍次克海域沉积物样品(lv39-06-06-04-18H)分离得到的150株低温细菌中筛选到菌株130产脂肪酶活性最高,其活性达到42 U/mL。对该菌株进行16S rRNA基因序列同源性和系统发育分析,结果表明菌株130属于芽孢杆菌属(Bacillus)。对其分泌脂肪酶的部分酶活特性进行研究,结果表明:酶的最适作用温度为35℃,0℃时保持33%的相对酶活力;对热敏感,60℃处理10 min仅残留30%酶活性;酶的最适pH范围在6.0~7.0。

【Abstract】 Low-temperature lipase producing bacterial strain 130 was screened from 150 isolates from Okhotsk Sea sediments samples(lv39-06-06-04-18H),and its activity reached 42 U/mL.The 16S rRNA gene sequences homology and phylogenetic analysis showed that strain 130 belonged to the genus Bacillus.The lipase characteristic had been studied,and the result showed that the optimal temperature of the action of this lipase was 35 ℃ and about 33 % of the optimum activity was still detectable at 0 ℃.The enzyme was thermal lability,only 30 % of its activity was remained after 10 min incubation at 60 ℃.The enzyme showed high lipolytic activity from pH 6.0 to 7.0.
