山楂残次果果胶提取工艺的优化
Study on optimization of extraction technology for pectin from substandard haw fruit by response surface methodology
【摘要】 利用微波-盐酸-盐析法对山楂残次果果胶的提取工艺进行研究。采用单因素试验研究盐酸浓度、液料比、微波比功率、微波时间、盐析条件对果胶得率的影响,在单因素试验基础上,根据中心组合(Box-Behnken)试验设计原理设计3因素3水平的响应面分析法,依据回归分析各影响因素,建立二次多项式回归方程的预测模型。以果胶得率为响应值作响应面和等高线,得出山楂残次果果胶提取的最佳工艺条件为盐酸浓度0.10mol/L,微波比功率167W/g,液料比16∶1(V∶m),饱和硫酸铝溶液用量7mL,微波时间50s,在优化条件下果胶得率为6.17%。
【Abstract】 Pectin was extracted from substandard hawthorn fruit with microwave-hydrochloric acid-salting out.The single-factor tests were used to investigate the effects of hydrochloric acid concentration,liquid-material ratio,microwave output power per gram,microwave extraction irradiation time,condition of salting-out on the yield rate of pectin.The response surface method with 3 factors at 3 levels,which was based on single-factor tests,was designed to analyze the all of affecting factors,and a quadratic polynomial regression equation of the forecasting model was set up according to the principle of Box-Behnken experiment.The yield rate of pectin from substandard hawthorn being taken as the value for the response surface method and contour,the optimum extraction condition condition was confirmed as follows: the hydrochloric acid concentration,0.10 mol/L;the microwave output power per gram,167 W/g;the microwave time,50 s;the volume of saturated aluminum sulfate,7ml;and the liquid-material ratio,16∶1,under which condition,the yield rate of pectin was 6.17%.
