CXCL12通过PI3K/AKT信号通路对大鼠海马培养细胞乙酰胆碱分泌的影响

【摘要】 目的:观察CXCL12通过PI3K/AKT信号通路对体外培养的海马神经细胞乙酰胆碱合成和分泌的影响。方法:通过Western Blot方法观察CXCL12对培养的海马神经元PI3K/AKT信号通路和胆碱乙酰转移酶(ChAT)表达量的影响;ELISA方法分别检测CXCL12对海马神经细胞乙酰胆碱酯酶(AChE)的活性和乙酰胆碱分泌的影响,以及PI3K/AKT阻断剂(LY2940002)阻断海马培养神经元PI3K/AKT信号通路后乙酰胆碱的分泌和AChE活性的变化。结果:在海马神经细胞体外培养过程中,采用50 ng/ml的CXCL12作用于海马神经细胞后,AKT磷酸化水平升高,ChAT蛋白表达增多,AChE活性减少。随着培养时间的延长(1,3,5,7 d),乙酰胆碱的分泌逐渐增加;通过LY2940002阻断PI3K/AKT信号通路,AKT磷酸化水平降低,ChAT蛋白表达减少,AChE活性增加,乙酰胆碱分泌减少。结论:CXCL12可能通过PI3K/AKT信号通路引起乙酰胆碱合成和分泌增加,表明CX-CL12可以作为一种重要的神经生长因子刺激乙酰胆碱的合成和分泌。

【Abstract】 Objective: To explore the effects of CXCL12 on synthesis and secretion of acetycholine through PI3K/AKT pathway in rats.Methods: Effects of CXCL12 on the expression of choline acetyl transferase(ChAT) and PI3K/AKT signal pathway in cultured hippocampus cells were studied by Western Blot.In addition,the effects of CXCL12 on the activity of acetylcholinesterase(AChE) and the secretion of acetycholine(Ach),the change of culture cells secreting ACh and AChE activity after using PI3K/AKT blocking agent-LY294002 were detected by ELISA method,respectively.Results: In the presence of 50 ng/ml CXCL12 in cultured hippocampus cells,the level of phosphorylation of AKT and expression of ChAT were raise,and the AChE activity decreased.Following the culture time prolongation(1,3,5,7 d),the secretion of acetycholine gradually was increased.After the PI3K/AKT signal pathway in cultured hippocampus cells was blocked by LY294002,the level of phosphorylation of AKT and expression of ChAT sharply decreased,the AChE activity increase and the secretion of acetycholine reduced.Conclusion: CXCL12 may be involved in acetycholin synthesis,secretion through PI3K/AKT pathway and indicate CXCL12 can provoke acetycholin synthesis,secretion as one pivotal nerve growth factor.
