【摘要】 目的探讨白细胞介素-27(IL-27)基因多态性与鼻咽癌易感性的关系。方法以190例鼻咽癌患者和200例健康对照者为研究对象,应用聚合酶链反应—限制性片段长度多态性和DNA测序的方法对二者IL-27基因-964 A/G和4730 T/C单核苷酸多态性进行基因分型,同时采用酶联免疫吸附试验检测其血清IL-27水平。结果鼻咽癌组和正常对照组中,IL-27(-964)AA、AG、GG三种基因型的分布频率分别为47.4%和42.5%,41.1%和43.5%,11.6%和14.0%;IL-27(4730)TT、TC二种基因型的分布频率分别为82.6%和79.0%,17.4%和21.0%。IL-27基因型和等位基因在两组人群中的分布频率差异无统计学意义(P>0.05),-964 G等位基因携带的鼻咽癌患者血清IL-27水平显著低于不携带者(P<0.05)。结论 IL-27基因-964 A/G和4730 T/C多态性与鼻咽癌的易感性无关,但-964 G等位基因携带可能与鼻咽癌病情严重程度有关。更多还原

【Abstract】 Objective To investigate the association between genotypes polymorphism of IL-27 and the susceptibility of nasophayngeal carcinoma(NPC).Methods Two single nucleotide polymorphisms of IL-27 gene-964 A/G and 4730 T/C in 190 patients with NPC and 200 age and sex matched controls in a Chinese population were analyzed using a polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) strategy and DNA sequencing,and the serum level of IL-27 was determined by enzyme-linked immunosorbent assay(ELISA).Results The frequencies of AA,AG and GG genotype were 47.4%,41.1%,11.6% in NPC patients respectively,while in controls were 42.5%,43.5%,14.0%,respectively.The frequencies of TT and TC genotype were 82.6% and 17.4% in NPC patients respectively,while in controls were 79.0% and 21.0%.The frequencies of IL-27 genotypes and alleles were not significantly different between NPC and control groups(P>0.05).the serum level of IL-27-964 G allele carriers was significantly lower than no carriers in patients with NPC(P<0.05).Conclusions The polymorphisms of IL-27 gene-964 A/G and 4730 T/C associate with the susceptibility of NPC,but IL-27 gene-964 G allele may related with the risk of NPC.更多还原
