ERK-1/2对N/OFQ调节大鼠皮层神经元延迟整流钾电流激活动力学的影响
Effects of ERK-1/2 on the modulation of delayed rectifier potassium current activation kinesics by N/OFQ in acutely dissociated rat cerebral parietal cortical neurons
【摘要】 目的研究ERK-1/2对N/OFQ调节大鼠皮层神经元延迟整流钾电流(IK)激活动力学的影响,初步探讨N/OFQ调节大鼠皮层神经元IK的ERK-1/2途径。方法研究采用全细胞膜片钳技术,观察ERK-1/2特异性抑制剂U0126对N/OFQ调节大鼠皮层神经元IK激活动力学的影响。结果在ERK-1/2抑制剂U0126存在情况下,IK激活曲线的V1/2由给药前的(-42±3.7)mV向超极化方向移动为(-39.9±5.9)mV(P<0.01)。IK激活曲线的κ由给药前的(21.6±8.1)mV变为给药后的(22.9±3.8)mV(P<0.05)。结论 U0126可以显著抑制N/OFQ对IK激活曲线的影响。
【Abstract】 Objective To investigate the effects of ERK-1/2 on modulation of delayed rectifier potassium current(IK) activation kinesics by N/OFQ in acutely dissociated rat cerebral parietal cortical neurons.Method Whole-cell voltage clamp technique was used.Result In presence of U0126,inhibitor of ERK-1/2,0.1 μmol/L N/OFQ shifted the activation curve of IK to hyperpolarize potential from(-42 ± 3.7) mV to(-39.9 ± 5.9) mV(P<0.01) and changed the slope factor(κ) of the activation curve from(21.6 ± 8.1) mV to(22.9 ± 3.8) mV(P<0.05).Conclusion These data sustained our contention that U0126 inhibited the positive shift of IK activation induced by N/OFQ.
