黄芩苷对巨噬细胞产生抵抗素和白细胞介素-1β的影响
Effect of Baicalin on Production of Resistin and Interleukin 1β in Macrophages Induced by Lipopolysaccharide
【摘要】 【目的】观察黄芩苷对脂多糖(LPS)诱导小鼠巨噬细胞产生抵抗素和白细胞介素-1β(IL-1β)的影响。【方法】体外培养小鼠巨噬细胞RAW 264.7,取生长良好的细胞进行接种。模型组加入LPS(终浓度为0.1μg/mL或5μg/mL)或重组抵抗素(终浓度为100 ng/mL);中药组加入不同浓度黄芩苷(终浓度为50、100μmol/L),预处理1 h,后加入LPS或抵抗素。24 h后吸取上清,采用酶联免疫吸附(ELISA)法检测各组IL-1β和抵抗素浓度,提取细胞总RNA,采用实时荧光定量PCR法检测抵抗素mRNA表达。【结果】与正常组比较,LPS组促进了巨噬细胞分泌抵抗素,抵抗素mRNA表达上调,LPS和重组抵抗素均促进了巨噬细胞产生IL-1β(P<0.05或P<0.01)。黄芩苷高、低剂量组均可显著抑制IL-1β和抵抗素的浓度,且抑制抵抗素mRNA表达(P<0.05或P<0.01)。【结论】黄芩苷可明显降低促炎因子产生的炎症反应,这可能是其抗动脉粥样硬化的机制之一。
【Abstract】 Objective To study the influence of baicalin on resistin and interleukin 1β(IL-1β) production in macrophages induced by lipopolysaccharide(LPS).Methods Well-grown mice macrophage(RAW264.7 cells) cultured in vitro were divided into normal control group,model groups(added with LPS at final concentration of 0.1 μg/mL or 5μg/mL,or with recombinant resistin at the concentration of 100 ng/mL),Chinese medicine groups(pretreated with baicalin at the concentration of 50 or 100 μmol/mL for one hour,and then added with LPS or recombinant resistin).Twenty-four hour later,the supernatant of the culture liquid was prepared for the detection of IL-1β and resistin concentrations with enzyme linked immunosorbent assay(ELISA).Total RNA was extracted from RAW264.7 cells,and resistin mRNA expression in the cells was detected by real-time fluorescence quantitative PCR method.Results Compared with the normal control group,LPS promoted the secretion of resistin in the macrophages,and correspondingly resistin mRNA expression was increased,and both LPS and recombinant resistin promoted the production of IL-1β in macrophages(P<0.05 or P<0.01).High-and low-dosage baicalin markedly decreased IL-1β and resistin contents as well as resistin mRNA expression(P<0.05 or P<0.01 compared with those in the model groups).Conclusion Baicalin can obviously reduce inflammatory reaction caused by proinflammatory factors,which may be one of its anti-atherosclerosis mechanisms.
