【摘要】 目的:探讨CXCR4RNA干扰对结肠癌细胞株SW480CXCR4mRNA和CXCR4蛋白凋亡、迁移和侵袭能力的影响。方法:采用脂质体转导CXCR4RNA干扰序列sihCXCR4-3,RT-PCR检测CXCR4mRNA相对含量,蛋白质印迹法检测CXCR4蛋白表达水平,流式细胞术检测细胞凋亡,Transwell小室评价SW480细胞迁移与侵袭能力。结果:sih-CXCR4-3组CXCR4mRNA的抑制率为81.40%,CXCR4蛋白相对含量为9.62%。早期凋亡率各组间均无差别,晚期凋亡率sihCXCR4-3组为(7.62±0.88)%,高于SW480细胞组(6.41±0.26)%,P=0.045 1。迁移实验显示,sihCXCR4-3组平均细胞数为84.25±22.46,低于SW480组(154.63±44.53,P=0.002 6)与NCsihCXCR4组(110.63±20.16,P=0.026 6)。侵袭实验发现,sihCXCR4-3组平均细胞数为0,低于SW480组(294.43±70.10,P=0.0000)与NCsihCXCR4(201.43±54.41,P=0.0000)。结论:CXCR4RNA干扰抑制SW480细胞CXCR4基因与蛋白表达,促进凋亡,降低迁移和侵袭能力。更多还原

【Abstract】 OBJECTIVE:To investigate the influence of CXCR4 RNA interference to apoptosis,migration,and invasion of SW480 cell.METHODS:Lipoplast was employed to transduce CXCR4 RNA interference sequence sihCXCR4-3,RT-PCR and Western blot were used to identify the ideal RNA interference effectiveness,flow cytometry was used to detect apoptosis,and Transwell chamber was applied to evaluate the migration and invasion of SW480 cell.RESULTS:The inhibition ratio of CXCR4 mRNA was 81.40%,and relative expression of CXCR4 protein was 9.62% in sihCXCR4-3 group.The early apoptosis was equal among three groups.Late apoptosis was higher in sihCXCR4-3 group than that in SW480 cell group((7.62±0.88)% vs(6.41±0.26)%,respectively,P=0.045 1).The migration experiment showed the mean number of SW480 cell in sihCXCR4-3 group(84.25±22.46) was lower than that in SW480 group(154.63±44.53,P=0.002 6)and NCsihCXCR4 group(110.63±20.16,P=0.026 6).The invasion experiment showed the mean number of SW480 cell in sihCXCR4-3 group(0) was lower than that in SW480 group(294.43±70.10,P=0.000 0)and NCsihCXCR4 group(201.43±54.41,P=0.000 0).CONCLUSION: CXCR4 RNA interference inhibits CXCR4 gene expression,promotes apoptosis,and degrades the migration and invasion power of SW480 cell.更多还原
