榄香烯对恶性淋巴瘤细胞多药耐药性影响及机制的研究
Influence of clemene on MDR of the lymphoma cell and it’s mechanism
【摘要】 目的:研究榄香烯对恶性淋巴细胞瘤细胞多药耐药性的影响及相关机制。方法:以不同浓度榄香烯作用于恶性淋巴瘤Raji细胞,并设空白对照组。通过RT-PCR以及蛋白质印迹方法检测加药后Raji细胞环氧化酶-2(COX-2)、核因子-κB(NF-κB)的mRNA及COX-2、NF-κB 56蛋白表达情况。结果:24h后,RT-PCR产物经电泳分析,COX-2mRNA、NF-κB mRNA各扩增产物在其相应位置上可见清晰的条带,特异性好。COX-2mRNA、NF-κB mRNA灰度值,各浓度榄香烯处理组与阴性对照组相比差异有统计学意义,P<0.01。各浓度榄香烯处理组与阴性对照组不同程度地表达COX-2、NF-κB p65蛋白,各浓度榄香烯处理组COX-2、NF-κB p65蛋白表达量与阴性对照组相比均下降,呈浓度依赖性,P<0.01。结论:榄香烯可能通过对COX-2和NF-κB的调控影响恶性淋巴细胞瘤细胞多药耐药性。
【Abstract】 OBJECTIVE: To explore the influence of clemene on the expression of COX-2,NF-κB in Raji cell and it’s mechanism.METHODS: The different doeses of clemene were used on Raji cell,comparing to another five contrasting medicine.Experssion of mRNA of COX-2,NF-κB and COX-2,NF-κB p56 proteinum were determined through RT-PCR and Western blot methods.RESULTS: Suppression of all medicines on the expression of mRNA of COX-2,NF-κB after 24 h.There was a difference betweent the different does clemene on the expression of COX-2,NF-κB and 0 μmol/L groups(P<0.01).Western blot results showed that a specific protein strip of COX-2 and NF-κB p56 expression was observed at the 72×103 and become more obviously following does(P<0.01).CONCLUSION: The clemene has effect on the expression of COX-2 and NF-κB,and decase the MDR of Raji cell.
