结直肠癌组织胸苷磷酸化酶表达临床意义研究的现状

【摘要】 目的:总结近年来关于胸苷磷酸化酶(TP)在结直肠癌组织中表达临床意义的研究现状。方法:应用PubMed、Elsevier Sciencedirect、CNKI及万方全文数据库检索系统,以胸苷磷酸化酶和结直肠癌为关键词,检索2005-01-2011-06发表的相关文献。纳入标准:1)TP在结直肠癌及转移组织中的表达及其促血管新生、抗细胞凋亡等生物学作用。2)TP与5-氟尿嘧啶(5-FU)前体类药物靶向治疗的关系。根据纳入标准分析文献29篇。结果:TP对于结直肠癌的影响是双重性的,其在肿瘤血管新生、转移、复发以及抗细胞凋亡等过程中均发挥重要作用,高表达TP的患者往往预后不良。另外,TP又在5-FU前体药物的化疗过程中起着关键作用,增加TP的表达很可能可以增强5-FU前体药物的化疗效果。结论:TP在结直肠癌组织中的表达对结直肠癌靶向治疗和预后有着非常重要的临床意义。

【Abstract】 OBJECTIVE:To summarize the advances of TP in colorectal cancer in recent years.METHODS: Using thymidine phosphorylase and colorectal cancer as Key words,the articles published from the database such as PubMed,Elsevier Sciencedirect,CNKI and Wangfang,particularly from 2005-01 to 2011-06 were searched.Inclusion Criteria: 1) The expression of TP in colorectal cancer and metastatic tissues and TP played an important role in the process of tumor angiogenesis,anti-apoptosis etc.2) The relationship between TP and targeted therapy of prodrugs of 5-fluorouracil(5-FU).Twenty-nine articles were selected into analysis according to inclusion criteria.RESULTS: TP has dual impact on colorectal cancer.It plays an important role in the process of tumor angiogenesis,metastasis,palindromia,and anti-apoptosis.The patients had high level of TP tend to have bad prognosis.TP is closely related to sensibility of tumor cells to prodrugs of 5-FU.Increasing the expression of TP is likely to enhance the effect of chemotherapy of prodrugs of 5-FU.CONCLUSION: The expression of TP in colorectal cancer and metastatic tissues has extraordinary important clinical significance to argeted therapy and prognosis.
