Notch1胞内段在鼻咽癌组织及鼻咽癌细胞株中的表达研究

【摘要】 目的探讨Notch1胞内段(NIC)在非角化性鼻咽癌组织和细胞株中的表达。方法采用反转录聚合酶链反应(RT-PCR)及免疫组化染色方法对鼻咽癌标本及不同分化程度鼻咽癌细胞株进行检测。结果 RT-PCR检测显示不同鼻咽癌细胞株中NIC基因均有表达,但分化程度高的CNE1细胞株表达最明显;同期进行的免疫组化染色结果与之相似;对鼻咽癌标本的免疫组化染色结果显示:鼻咽癌组织中NIC的表达高于鼻咽慢性黏膜炎症组织(P=0.005),而且NIC的表达强度与细胞分化程度密切相关(P<0.05),与患者性别、年龄无相关性(P>0.05)。另外,在未分化型鼻咽癌中,癌巢内几乎无阳性表达,淋巴细胞阳性表达多;而在分化型鼻咽癌中癌细胞阳性表达明显,可见于胞膜、胞质及胞核。结论 Notch信号系统参与鼻咽癌的发生,其中Notch1的表达与鼻咽癌的分化程度相关。

【Abstract】 Objectives To investigate the expression of Notch1 intracellular domain(NIC)in non-kertain nasopharyngeal carcinoma(NK-NPC).Methods RT-PCR and immunocytochemical staining were used to determine 52 biopsy specimens and different cell-lines of NPC.Results By RT-PCR,NIC genes were expressed in different NK-NPC cell lines,but the expression of high-differentiated CNE1 cell line was most remarkable.The results of immunocytochemical staining performed correspondingly were similar to it.By the results of immunocytochemical staining of NPC specimens,NIC expression was higher in NK-NPC than in chronic nasopharyngitis(P=0.005),closely correlated with differentiation(P<0.05),but not with gender or age(P>0.05).In undifferentiated NK-NPC,there was scarcely positive expression but more lymphocyte positive expression.In differentiated NK-NPC,positive expression was obvious and cell membrance,periplasm and nucleus were seen.Conclusion Notch signaling participates in occurance of non-keratin naspharyngeal carcinoma,and Notch1 expression is positively correlated with differentiated degree of NK-NPC.
