化痰祛湿剂对兔膝骨性关节炎软骨及滑膜的影响
Effect of Huatan Qushi Preparation Xibikang on the Ppathologic Change of Cartilage and Synovial Membrane of Rabbit Knee Osteoarthritis
【摘要】 目的:观察化痰祛湿剂膝痹康对兔实验性膝骨性关节炎模型关节软骨及滑膜病理学变化的影响。方法:健康家兔40只,体重2.8~3.2kg,随机分为模型组、膝痹康组(为本院制剂,批号050411,主要由制南星、制川乌、薏苡仁、刺五加、丹参、地龙等组成,含生药2.0g/mL的合剂,功效化痰祛湿、消肿止痛)、阳性药(美洛昔康)对照组、假手术对照组,每组家兔10只。模型组、膝痹康组、阳性药(美洛昔康)对照组家兔通过改良的Hulth法复制兔膝关节炎动物模型,假手术组仅切开膝关节表面的皮肤,膝痹康组、阳性药对照组家兔于造模成功后开始用药,连续用药8周,4组家兔于造模后全部处死,切除患侧胫骨全层关节软骨及滑膜进行肉眼、光镜组织学检查,并对关节软骨进行透射电镜检查。结果:显示模型组兔患侧关节软骨表面粗糙,变暗呈灰黄色,并有裂纹、糜烂及溃疡形成;滑膜存在不同程度增生、粘连,关节液量增多,且呈泡沫状、浑浊,Mankin’s评分显著高于假手术对照组;膝痹康组关节软骨及滑膜虽未能恢复到正常状态,但Mankin’s评分较模型组显著降低。结论:膝痹康可明显减轻膝骨性关节炎兔关节软骨及滑膜的病理损伤,对软骨及滑膜的退变有较好的延缓趋势。
【Abstract】 Objective:To observe the effects of Huatan Qushi preparation xibikang on the pathologic change of cartilage and synovial membrane of rabbit knee osteoarthritis.Methods:40 healthy rabbits were equally devided 4 groups at random.Using improved Hulth procedure established animal models of rabbit knee osteoarthritis.Those rabbits were taken xibikang by mouth 8 weeks.Cutting the full-thickness cartilage and synovial membrane of those rabbits’ affected tibia,the tissue was observed by naked eyes and light microscope 30 minutes after the last time taking medicine.Transmission electron microscope(TEM)was applied to observe cartilage.Results:The results showed that the rabbits’ affected cartilage was rough and isabeling,crack,erosion and ulcer were formed;the synovial membrane appeared hyperplasia and adhere;synovia was increased and appeared cystose and muddy;the Mankin’s score were notablely higher than false operation group;xibikang group’s cartilage and synovial membrane didn’t cecover but the Mankin’s score were notablely lower than model group.Conclusion:Suggesting,xibikang can obviously reduce the pathological injury of cartilage and synovial membrane in rabbit osteoarthritis and can delay the cataplasia of cartilage and synovial membrane.
