FK506作用下肠黏膜屏障功能的变化及意义

【摘要】 目的:探讨FK506作用下肠屏障功能的变化及意义。方法:比较免疫抑制组和对照组大鼠血浆D-乳酸和DAO浓度、回肠黏膜病理分析。结果:免疫抑制组血浆D-乳酸和DAO浓度、肠黏膜损伤评分在各时间点均呈缓慢上升趋势。与对照组比较,第3、5、7天血浆D-乳酸和DAO浓度、各时间点的肠黏膜损伤评分均显著增高(P<0.05)。血浆D-乳酸、DAO浓度与肠黏膜损伤评分之间呈显著正相关(P<0.05)。结论:FK506可以引起肠黏膜上皮损伤,肠黏膜机械屏障功能破坏,肠黏膜通透性增高。随FK506应用时间的延长,肠黏膜损伤的加重,肠黏膜通透性改变越显著。检测血浆D-乳酸和DAO的变化,能够及时反映免疫抑制剂引起的肠黏膜上皮损伤及其通透性变化。
【Abstract】 Objective:To investingate the changes of gut barrier function under the effects of FK506.Methods:Concentration of D-lactic and DAO in blood,intestinal mucosal lesion scores between immunosuppression group and control group rats were compared.Results:Concentration of D-lactic and DAO in blood,intestinal mucosal lesion score were on the rise slowly at all time points in immunosuppression group rats,respectively.Compared with control group,concentration of D-lactic and DAO in blood at 3th,5th,7th day,intestinal mucosal lesion score at all time points were higher significantly in immunosuppression group,respectively(P<0.05).Concentration of D-lactic and DAO in blood were related significantly to intestinal mucosal lesion score,respectively(P<0.05).Conclutions:FK506 could cause damage to intestinal epithelia,disrupt intestinal mechanical barrier,increase intestinal permeability.Furthermore,with the time lenthening of using immunosuppressants and the aggravating of intestinal mucosal injury,the intestinal mucosal permeability changes more significantly.It maybe one of the main reasons that the use of large dose immunosuppressants can cause gut-derived infection.Detecting of D-lactic and DAO in blood could reflect damage of intestinal mucosal epithelium and change of gut permeability under immunosuppressants in time.
