淫羊藿有效单体对活化的肾成纤维细胞株和系膜细胞株的影响
The Effect of Utility Monomer in Epimedium to the Renal Fibroblast Cell Line and Mesangial Cell Line
【摘要】 目的:观察淫羊藿有效单体对活化的肾成纤维细胞株和系膜细胞株的影响。方法:采用体外培养肾成纤维细胞株和系膜细胞株的方法,将其分为空白对照组、TGF-β12ng/ml组、TGF-β12ng/ml+中药10-5g/ml组和TGF-β12ng/ml+中药10-6g/ml组。采用MTT法观察24h中药单体对两种细胞株的作用以及ELISA法对TGF-β的分泌情况。结果:与空白对照组相比,加入TGF-β1刺激因子可以促进两种细胞株的活化(P<0.05),以及TGF-β的分泌(P<0.01),中药单体可以抑制活化的细胞株增殖和TGF-β的分泌(P<0.05或P<0.01)。结论:淫羊藿中药单体可以抑制活化的肾成纤维细胞株和系膜细胞株的增殖,有效防治肾纤维化,其作用机制可能与其抑制TGF-β的分泌有关。
【Abstract】 Objective:To observe the effect of utility monomer in Epimedium to the renal fibroblast cell line and mesangial cell line.Methods:To culture in vitro the renal fibroblast cell line and mesangial cell line,were divided into blank control group,TGF-β1 2 ng/ml group,TGF-β1 2 ng/ml+Chinese medicine monomer 10-5 g/ml group and TGF-β1 2 ng/ml + Chinese medicine monomer 10-6 g/ml group.To observe the effect of Chinese medicine monomer for 24 hours the two cell lines by MTT assay,and the secretion of TGF-β by ELISA assay.Results:Compared with blank control group,adding TGF-β1-stimulating factor can promote the two cell lines (P<0.05),as well as the secretion of TGF-β (P<0.01),Chinese medicine monomer can inhibit the activation of two cell lines proliferation and secretion of TGF-β (P<0.05 or P<0.01).Conclusion:Utility monomer of Epimedium can inhibit the activation of the renal fibroblast cell line and mesangial cell line proliferation and secretion of TGF-β,effective prevention and treatment of renal fibrosis,and its possible mechanism of action relate to inhibition of TGF-β secretion.
